摘  要:目的探讨热休克蛋白70(HSP70)反义脱氧寡核苷酸(ASODN)对体外培养的人卵巢癌细胞株HO-8910生长和凋亡的影响。方法取对数生长期HO-8910人卵巢癌细胞,与不同终浓度(5、10、15和20μmol/L)的HSP70 ASODN及正义脱氧寡核苷酸(SODN)共同孵育。四甲基偶氮唑蓝光吸收法(MTT)检测细胞增殖情况;流式细胞仪分析细胞DNA含量及细胞周期动力学改变。结果MTT检测表明HSP70 ASODN能够抑制HO-8910细胞增殖,且一定范围内呈浓度、时间依赖性。流式细胞仪检测分析可见亚二倍体峰,凋亡指数(AI)随HSP70 ASODN浓度升高而升高,差异有统计学意义(P<0.05)。细胞周期分析提示HSP70 ASODN使细胞停滞于G1/G0期,S期细胞逐渐减少,G2/M期无明显变化。结论HSP70 ASODN可抑制卵巢癌细胞HO-8910生长并可诱导凋亡。Objective To investigate the effect of heat shock protein 70 (HSP70) antisense oligodeoxynucleotides (ASODN) on growth and apoptosis of ovarian cancer cells line HO-8910. Methods Human ovarian cancer cell line HO-8910 was treated in vitro with various concentrations(5, 10, 15 and 20 μmol/L)of HSP70 ASODN and sense oligodeoxynucleotides (SODN) at different intervals. The growth inhibiting rates of HO-8910 cells were assayed by methyl thiazolyl tetrazolium(MTT) , and the DNA content and cell cycle were analysed by flow cytometer. Results MTT assay showed that HSP70 ASODN could inhibite the growth of ovarian cancer cells depending on the dose and time of HSP70 ASODN in some range. Flow cytometry analysis showed a suhdiploidy cell peak. Apoptotic index (AI) in HSP70 ASODN group increased with the increase of concentration of HSP70 ASODN. Cell cycle indicated the decrease in the percentage of cells in the G1 and S phases, but the cells in the G2 phases of the cell cycle had no obvious change. Conclusions HSP70 ASODN can inhibite growth and induce apoptosis of HO-8910 cells.
