摘 要:目的建立测定通舒口爽胶囊中大黄素、大黄酸、大黄酚和大黄素甲醚含量的HPLC方法。方法采用Diamonsil C18(200mm×4.6mm,5μm)色谱柱,以甲醇0.05%磷酸水溶液(92:8,v/v)为流动相,流速:1.0mL·min^-1,柱温:30℃,检测波长:254nm。结果大黄素在0.7~7.0μg·mL^-1呈线性关系(r=0.9999),平均回收率为99.3%;大黄酸在0.384-3.84μg·mL^-1呈线性关系(r=0.9996),平均回收率为100.7%;大黄酚在1.803~18.03μg·mL^-1呈线性关系(r=0.9997),平均回收率为100.9%;大黄素甲醚在0.504-5.04μg·mL^-1呈线性关系(r=0.9999),平均回收率为99.1%。结论本方法操作简便、可靠、重现性好、专属性好,可作为通舒口爽胶囊质量控制方法之一。Objective To develop an HPLC method for the determination of emodin, rhein, chrysophanol and rheoehrysidin in Tongshukoushuang capsules. Methods Diamonsil C18 (200 mm ×4. 6 mm, 5 μm) column was used with the mobile phase of methanol-0. 05% phosphoric acid solution (92 : 8, v/v) . The flow rate was 1.0 mL · min^-1 and the column temperature was 30 ℃. The detection wave-length was 254 nm. Results The calibration curve was linear within 0.7-7.0 μg · mL^-1 (r=0. 999 9), 0. 384--3.84μg · mL^-1(r= 0. 999 6), 1. 803-18.03μg · mL^-1 (r= 0. 999 7), and 0. 504-5.04 μg · mL^-1(r=0. 999 9) for emodin, rhein, chrysophanol, and rheochrysidin, respectively. The average recoveries were 99. 3%, 100. 7%, 100. 9% and 99.1% for emodin, rhein, chrysophanol, and rheochrysidin, respectively. Conclusion This method is convenient, accurate and reproducible for the determination of emodin, rhein, chrysophanol, and rheoehrysidin in Tongshukoushuang capsules.
