摘  要:目的探讨静脉输注同种异基因大鼠骨髓间质干细胞(MSCs)对体内细胞免疫功能的影响。方法从Wistar大鼠骨髓分离培养MSCs,通过形态学观察及流式细胞术检测表面标志以鉴定其纯度。选择20只正常SD大鼠,随机均分为4个组,分别经静脉输注5×106/ml、5×105/ml、5×104/ml的MSCs及PBS各1ml。10d后取受体SD大鼠脾脏行混合淋巴细胞反应(MLR)检测淋巴细胞增殖率、外周血CD4+、CD8+、CD4+/CD8+及CD4+CD25+/CD4+T淋巴细胞比率,以评价体内MSCs对正常大鼠细胞免疫功能的影响。结果输注MSCs达5×106/ml的受体SD大鼠脾脏淋巴细胞增殖率为8.58%±0.27%,明显低于输注PBS大鼠(24.40%±5.21%,P<0.01)。输注5×106/ml MSCs的SD大鼠脾脏、外周血CD4+下降,CD4+CD25+/CD4+T细胞比率上升,与注射PBS大鼠相比差异有统计学意义(P<0.01)。输注5×105/ml、5×104/ml MSCs的大鼠各项指标与输注PBS大鼠相比差异无统计学意义(P>0.05)。结论高浓度同种异基因MSCs静脉输入后可以部分抑制活体的细胞免疫功能,提高CD4+CD25+/CD4+T细胞的比率,为诱导移植免疫耐受及治疗自身免疫疾病提供了理论基础。Objective To investigate the effects of injecting allogeneic mesenchymal stem cells (MSCs) on the cellular immunity in rat invivo. Methods Bone marrow-derived MSCs were isolated from Wistar rats. The purity of MSCs was identified by morphological exarnination with microphotography, and the phenotypes were identified with flow cytometry. Twenty SD rats were randomly divided into 4 groups. Different concentrations of MSCs (5×10^6/ml for group A, 5×10^5/ml for group B, and 5×10^4/ml for group C, respectively) and PBS (for group D) were given to allogeneic SD rats via intravenous infusions. The suppressive effect of MSCs on lymphocytes proliferation in recipient rat was analyzed using mixed lymphocyte cultures (MLR) 10 days after cultivation. At the same time, proportions of CD4^+ , CD8^+ and CD4^+ CD25^+/CD4^+ T-lymphocytes in peripheral blood and spleen were analyzed with flow cytometry. Results Proliferation rate of splenic lymphoeytes in group A (5×10^6/ml MSCs, 8. 58%±0. 27%) was markedly lowered compared with that in group D (PBS, 24. 40%±5. 21%, P〈0. 01). The proportions of CD4^+ T-lymphocytes in spleen and periphery blood were decreased, and of CD4^+ CD25^+/CD4^+ T-lymphocytes increased in group A compared with that in group D (P〈0. 01). No significant difference was found when comparing the indexes mentioned above in groups B and C with that in group D. Conclusions Injection of allogeneic MSCs in vivo may influence the proliferation of lymphocytes. Moreover, cellular immunity may he suppressed after infusing MSCs in high-dosage. MSCs can increase the proportion of CD4^+ CD25^+/CD4^+ T lymphoeytes. These data have contributed a theoretical basis for the treatment of autoimmune diseases and host versus graft reaction in organ transplantation.
