摘  要:为研究过氧化氢对小麦洛夫林10悬浮细胞的影响,分别采用溴酚兰染色、改良苯酚品红染色和琼脂糖凝胶电泳对不同浓度的过氧化氢处理小麦悬浮细胞后的细胞死亡率、细胞核形态变化及基因组DNA的完整性进行了观察和检测。结果表明:随着过氧化氢浓度的增加以及处理时间的延长细胞死亡率逐渐上升,并且伴随有细胞核染色质的凝集,琼脂糖凝胶电泳检测到DNA Ladder。证明过氧化氢能够诱导小麦悬浮细胞发生程序性死亡。In order to study the effects of H2O2 on the suspension cells of wheat (Lovrin 10), the death rate, the changes of karyomorphology and the integrity of genomic DNA induced by different concentration of H2O2 were examined using bromphenol blue staining, improved phenol fuchsin staining and agarose gel electrophoresis. The results showed that the death rate of cells was gradually increased when the concentration of H2O2 was increased and the time after the treatment of H2O2 prolonged. At the same time the highly condensed chromatin in the cell nucleus accumulated during this process, and DNA ladder was detected by agarose gel electrophoresis. It can be summed up that H2O2 may induce programmed cell death of wheat suspension cells.
