云南烟草上出现番茄斑萎病毒(Tospovirus)属病毒  

Identification of tospovirus on tobaccos in Yunan province

摘　　要：2004-2007年间对云南省烤烟种植区进行调查，在云南省楚雄和文山2个地区，获得3份疑似Tospovirus属病毒的样本。通过Tospovirus组引物（Tospovirus Group Primer）进行RT-PCR扩增、克隆和测序，获得500nts核酸片段，通过NCBI BLAST网络数据库比对，确定这3份样品为Tospovirus属的病毒；通过DNAman软件比对分析所得到的核酸序列和已经公布的Tospovirus属病毒RNA L同位点序列，从得到的系统进化图可以看出，云南3个样品明显分为2簇，即YN-1和YN-2，分别与血清组I和Ⅵ的Tospovirus属病毒聚为一簇。Three samples from the genus Tospovirus were collected from major tobacco fields of Chuxiong and Wenshan in Yuannan from 2004-2007 survey. After RT-PCR, clone, sequencing using Tospovirus Group Primers and comparison with results from NCBI Blast, the 3 samples were confirmed to be from Tospovirus genus because of containing the Tospovirus RNA L fragments. A phylogenetic tree based on the sequence of three sample virus and their corresponding public RNA L fragment sequences of Tospovirus was constructed using DNAman software. The phylogenetic tree indicated that 3 Yuannan samples were grouped into two clusters： YN-1 and YN-2, which fell into the same cluster as the Serogroup I, and Ⅵ, respectively.
