摘  要:目的研究低浓度N-甲基-N′-硝基-N-甲基亚硝基胍对人羊膜FL细胞的细胞周期基因表达的影响,以助于阐明MNNG引起细胞应答反应的机制。方法用实时荧光定量PCR方法,检测FL细胞在1.0μmol/LMNNG处理后细胞周期基因CyclinE1及CyclinE2基因表达的改变,数据用ABI公司的SDS2.1软件分析。结果MNNG处理后,细胞周期基因CyclinE1及CyclinE2表达显著上调(P<0.05)。结论低浓度MNNG攻击可使人羊膜FL细胞周期发生异常。Objective To study the expression changes of genes associated with cell cycle in human FL cells with treatment of low concentration of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in order to elucidate the mechanism of response reaction of human FL cells exposed to MNNG. Methods CyclinE1 and CyclinE2 genes, associated with cell cycle, of human FL cells with or without treatment of 1.0 μmol/L MNNG were detected by real-time PCR of ABL Data were analyzed by SDS2.1 software. Results Results Expressions of CyclinE1 and CyclinE2 genes increased significantly after MNNG treatment (P 〈 0.05). Conclusion Low concentration of MNNG attack can result in abnormal cell cycle in human FL cells.
