低分子肝素对侵袭性肺曲霉病小鼠细胞因子的影响

摘  要:目的探讨低分子肝素(LWMH)对侵袭性肺曲霉病(IPA)小鼠血浆及肺泡灌洗液中肿瘤坏死因子(TNF)-α、白介素(IL)-1β和IL-10浓度的影响。方法采用环磷酰胺免疫抑制和烟曲霉孢子悬液滴鼻法(1×106个孢子/小鼠)构建中性粒细胞减少的小鼠IPA模型。120只小鼠随机分为四组(n=30):正常对照组(以生理盐水代替环磷酰胺和烟曲霉孢子悬液)、IPA模型组、生理盐水+LWMH组和IPA+LWMH组。生理盐水+LWMH组和IPA+LWMH组在滴鼻后即给予1000IU/kgLWMH,背部皮下注射,qd×2d;正常对照组与IPA模型组以等量生理盐水代替LWMH背部皮下注射。滴鼻后4、8、12、24、48h每组各随机取6只小鼠取血及肺泡灌洗液,ELISA法检测各时点血浆和肺泡灌洗液中TNF-α、IL-1β和IL-10浓度。结果IPA模型组与IPA+LWMH组小鼠接种烟曲霉孢子后血浆和肺泡灌洗液中TNF-α、IL-1β和IL-10浓度在数小时内即升高至较高水平;IPA+LWMH组较IPA模型组TNF-α和IL-1β浓度显著升高(P<0.05或P<0.01),而IL-10浓度差异无统计学意义(P>0.05);生理盐水+LWMH组与正常对照组这三种细胞因子浓度差异无统计学意义(P>0.05)。结论LWMH能显著提高IPA小鼠血浆及肺泡灌洗液中的TNF-α和IL-1β浓度。Objective To investigate the effects of low molecular heparin (LWMH) on cytokines (TNF-α, IL-1β and IL-10) in blood plasma and bronchoalveolar lavage fluid of invasive pulmonary aspergillosis (IPA) mice. Methods The neutropenic IPA mouse models were established by administration of cyclophosphamide for immunologic function inhibition and intranasally challenge with Aspergillus fumigatus conidia (1 × 10^6 conidia/mouse). One hundred and twenty mice were randomly divided into 4 groups : normal control, IPA model, normal saline + LWMH and IPA + LWMH group. Normal saline +LWMH group and IPA + LWMH group received LWMH (subcutaneous injection, 1 000 IU/kg, qd × 2 d). Normal control and IPA model group received normal saline instedad of LWMH. At 4,8,12,24 and 48 h after inoculation, six mice were randomly taken from each group to be sacrificed. ELISA method was used to determine the concentrations of TNF-α, IL-1β and IL-10 in blood plasma and BALF. Results TNF-α, IL-1β and IL-10 in blood plasma and BALF increased significantly several hours after inoculation of conidia in IPA model and IPA + LWMH group. There were significant higher concentrations of TNF-α and IL-1β in blood plasma and BALF in IPA + LWMH group than in IPA model group ( P 〈 0.05 or P 〈0.01) ,while there was no significant difference in IL-10 between the two groups. There was no significant difference in the concentrations of these cytokines between normal saline + LWMH group and normal control group. Conclusion LWMH can significantly increase the concentrations of TNF-α and IL-1β in blood plasma and BALF of IPA mice.
