摘  要:目的探讨过氧化物酶体增生物激活受体γ(PPARγ)激动剂罗格列酮(ROSI)对多器官功能障碍综合征(MODS)大鼠外周血单个核细胞(PBMC)内核因子-κB(NF-κB)表达的影响。方法健康雄性SD大鼠40只,随机分为4组(每组10只):①正常对照组;②LPS刺激组;③罗格列酮(ROSI)预处理组;④选择性拮抗剂2-氯-5-硝基苯胺(GW9662)预处理组。在每组操作结束后4h取血用于PBMC的分离。免疫细胞化学法检测PBMC内NF-κB p65的表达活性,并进行图像分析。结果在正常组大鼠PBMC内NF-κB p65表达较少。LPS刺激组NF-κB p65表达显著增加,与正常组比较差异显著(P<0.01)。ROSI组NF-κB p65表达减少,与LPS刺激组比较显著降低(P<0.01)。GW9662组NF-κB p65表达与LPS刺激组比较无显著差异性(P>0.05)。结论罗格列酮可能通过抑制NF-κB的表达活性而对MODS大鼠起保护作用。Objective To investigate the effects of rosiglitazone,an agonist of proliferators activated receptorγ(PPARγ) ,on expression of nuclear factor - kappa B ( NF -κB) in peripheral blood mononuclear cells ( PBMC ) of multiple organ dysfunction syndrome (MODS) rats. Methods Forty male SD rats were randomly divided into four groups (n = 6 per group) : vehicle control group, endotoxin (LPS) group,rosiglitazone(ROSI) pretreatment group, and PPAR -γantagonist GW9662 pretreatment group. Blood was taken 4 hours after operation, and mononuclear cells were separated. The expression of NF - κB p65in PBMC was detected by immunocytoehemieal method and image analysis was carried out. Results The expression of NF -κB p65 was low in vehicle control group. In LPS group, the expression of NF -κB p65 was significantly higher than that in vehicle control group( P 〈 0. 01 ). In ROSI group, the expression of NF -κB p65 was significantly lower than in that LPS group(P 〈0. 01 ). In GW9662 group, the expression of NF -κB p65 had no significant difference compared with LPS group(P 〉 0. 05). Conclusion Rosiglitazone protected the M ODS rats by inhibiting NF-κB activation.
