摘 要:目的:观察中药脑疏宁对创伤性脑损伤大鼠基质金属蛋白酶基质金属蛋白酶(MMPs)表达的影响。方法:采用免疫组化方法检测脑组织MMPs蛋白的表达,测定脑组织含水量及伊文思蓝(EB)含量,电镜下观察血脑屏障超微结构改变。结果:创伤组大鼠伤侧脑组织含水量、伊文思蓝(EB)含量及伤灶周围基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)的表达水平均明显高于假手术组(P<0.01);血脑屏障结构破坏。脑疏宁组脑组织含水量、EB含量及基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)的表达水平均明显低于创伤组(P<0.05,P<0.01);血脑屏障结构较为完整。结论:脑疏宁可通过改善血脑屏障损伤减轻创伤性脑水肿,其保护血脑屏障的作用可能是通过抑制MMP-2和MMP-9蛋白表达实现的。Objective: To explore the effect of Naoshuning on the protein expression of Matrix metalloproteinases (MMPs) in experimental injuried brain tissue of rats. Methods: Immunohistocbemistry was used to determine the changes of protein expression of MMPs. Brain tissue water content, permeability and ultramicrostructure of blood-brain barrier (BBB) were also observed. Results: Compared with the sham group, the brain tissue water and EB content of injured side and the level of MMP-2 and MMP-9 protein expression in brain tissue around contusion in model group increased obviously (all P〈0.01). The electron microscope result indicated that BBB was evidently damaged.In Naoshuning group, brain tissue water and EB content and the expression of MMP-2 and MMP-9 were obviously lower than those of the model group at each time point(P〈0.05-0.01). The ultramicrostructure changes could be relieved by Naoshuning. Conclusion: This study indicates that Chinese medicine Naoshuning could alleviate cerebral edema via protecting BBB, which might be realized by the inhibition on expression of MMP-2 and MMP-9 protein.
