摘  要:目的探讨补阳还五汤对老年性痴呆(AD)大鼠海马区β淀粉样前体蛋白(β-APP)及相关基因:环氧合酶2(COX-2)、核转录因子抑制蛋白α(IкB-α)、神经元型一氧化氮合酶(nNOS)的影响及可能的作用机制。方法采用Aβ1-40海马区注射,建立AD大鼠模型,免疫组化法观察中药补阳还五汤对AD大鼠海马区β-APP,COX-2,IкB-α及nNOS的影响。结果补阳还五汤能明显降低AD模型大鼠海马区β-APP,COX-2,IкB-α表达,使nNOS表达升高。结论补阳还五汤可能通过COX-2,NF-кB/IкB-α,增加模型大鼠海马区nNOS表达,影响β-APP生成,延缓神经元细胞的早期损伤和迟发性损伤,从而起到治疗AD的作用。Objective To observe the effects of Bu Yang Huan Wu Decoction for β-amyloid protein of AD rats caused by Aβ1-40, and immunologic mechanism. Methods AD rat model was established with Aβ1-40 by hippocampus injection, lmmunohistochemical method was used to detect the expression of β-APP, COX-2, nNOS and IKB-α in hippocampus of rats. Results Bu Yang Huan Wu Decoction obviously decreased the abnormal high expression of 13 - APP protein ;increased the masculine expression of nNOS, and decreased high masculine expression of COX - 2 and IKB -α in hippocampus of AD rats caused by Aβ. Conclusion Bu Yang Huan Wu Decoction might regulate the generation and metabolism of β - APP, reasonably control the immune inflammation cascade reaction caused by Aβ deposition through signal transduction pathway of COX - 2, IKB - α and nNOS. Therefore lessen brain neuronal damage effectually caused by Aβ toxic action ,and improve the dysmnesy.
