化痰祛瘀方对兔动脉内膜损伤后血管平滑肌细胞增殖的影响

摘　　要：目的探讨化痰祛瘀方对兔动脉内膜损伤后血管平滑肌细胞(VSMCs)增殖的影响。方法将30只新西兰兔随机分为正常组、模型组、氟伐他汀对照组及化痰祛瘀组。正常组6只,其余每组各8只。通过高脂饲料配合腹主动脉内膜剥脱术建立兔动脉内膜损伤模型。取腹主动脉下段,电镜观察血管平滑肌细胞形态;应用逆转录取聚合酶链式反应(RT-PCR)法测定原癌基因c-myc和血小板源性生长因子-BB(PDGF)水平;免疫组化法测定胰岛素样生长因子-1(IGF-1)水平。结果模型组PDGF、c-myc、IGF-1均显著高于正常组(P<0.01);氟伐他汀对照组、化痰祛瘀组与模型组相比较PDGF、c-myc、IGF-1显著下降(P<0.05)。结论动脉损伤后可致PDGF、c-myc、IGF-1表达增加,提示PDGF、c-myc、IGF-1表达增加在动脉损伤后平滑肌细胞增殖中起一定作用。化痰祛瘀方能使动脉损伤后PDGF、c-myc、IGF-1表达减少,对动脉损伤后平滑肌细胞增殖有一定抑制作用。Objective To investigate the effect of Huatanquyu decoction on vascular smooth muscle cells （VSMCs） proliferation in rabbits with endoarterium injury. Method Thirty New Zealand rabbits were randomly divided into the normal group （n=6）,model group （n=8）,fluvastatin contrast group （n=8） and Huatanquyu decoction group （n=8）. Rabbits endoarterium injured models were built by fed with high dosage of cholesterol diet with endoarterium denudation. Morphology of VSMCs was observed under the elec tron microscope. The level of pro oncogene c - myc and platelet derived growth factor - BB （PDGF - BB） were studied by RT - PCR and the level of insulin- like growth factor - 1 （IGF - 1） was studied by immunohistochemistry. Result Compared with the normal group, the level of PDGF - BB, c - myc and IGF - 1 were increased significantly in model group （P〈0.01）. Compared with the model group,the level of PDGF- BB,c- myc and IGF- 1 were decreased significantly in Huatanquyu decoction group and fluvastatin group （P〈0.05）. Conclusion After vascular intima injured,the level of PDGF - BB,c - myc and IGF - 1 were increased. Huatanquyu decoction could inhibit VSMCs proliferation by inhibiting expression of PDGF - BB,c - myc and IGF - 1.
