摘  要:目的观察抑郁对大鼠脑脊液、海马和皮层组织脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)的影响,探讨BDNF与抑郁症的发病关系及作用机制。方法采用慢性不可预见性应激法制作大鼠抑郁模型,运用Open-Field评分方法观察大鼠行为学变化;免疫组化法检测海马和皮层BDNF的表达,ELISA法检测脑脊液、海马和皮层BDNF的含量。结果与正常对照组比较,抑郁模型组大鼠海马和皮层BDNF阳性细胞数量均明显减少;正常对照组大鼠脑脊液、海马和皮层BDNF的含量分别为:(6.87±2.44)ng/ml、(11.75±3.60)ng/g、(4.31±1.37)ng/g,抑郁模型组大鼠脑脊液、海马和皮层BDNF的含量分别为:(4.23±1.54)ng/ml、(8.56±2.45)ng/g、(3.06±0.67)ng/g,表明抑郁模型组大鼠脑组织中BDNF的含量显著低于正常对照组(P<0.01)。结论抑郁模型大鼠脑组织中BDNF水平降低可能是导致抑郁症发生的重要因素之一。Objective To explore the effect of depression on the expression of brain-derived neurotrophic factor (BDNF) in the cerebral cortex, hippocampus and cerebrospinal fluid in order to investigate the relationship between BDNF and the occurrence of depression. Methods The depression model in rats was established with chronic unpredictable stress. The behaviors in Open-Field testing were observed through forced swimming. The expression of BDNF in above-mentioned tissues was examined by immunohistochemical method and ELISA. Results Compared with normal control, the number of BDNF positive cells in the hippocampus and cerebral cortex in depressed rats was decreased markedly. The level of BDNF in the cerebral-spinal fluid, hippocampus and cerebral cortex in normal mice was (6.87±2.44)ng/ml, (11.75 ±3.60)ng/g and (4.31± 1.37)ng/g respectively; and they were markedly higher than those of depressed rats [(4.23 ± 1.54)ng/ml, (8.56 ±2.45)ng/g and (3.06 ± 0.67)ng/g, P 〈 0.01 ]. Conclusion BDNF levels are obviously decreased in the depressed rats, indicating it may be involved in the occurrence of depression.
