摘  要:背景:研究证实丹参在创伤修复和愈合过程中起重要作用。滇丹参所含化学成分与正品丹参基本一致,其中某些活性成分含量高于正品丹参。目的:观察滇丹参对瘢痕成纤维细胞生物学行为的影响,揭示其对瘢痕的作用及机制。设计、时间及地点:分组对照观察,于2005-03/2006-03在昆明医学院完成。对象:瘢痕标本取自8例患者颌面颈部遗留瘢痕,男5例,女3例;年龄7~38岁;瘢痕病程1~12年,患者对实验均知情同意。方法:对瘢痕成纤维细胞进行体外培养,随机分组,分别加入0.05,0.1,0.2,0.4g/mL不同质量浓度的滇丹参液培养24h,阴性对照组加入不含滇丹参的DMEM培养液,阳性对照组加入含稀释成1000U/mLrhIFN-α2a的DMEM培养液。主要观察指标:采用光镜观察、四甲基偶氮唑盐比色测定分析、流式细胞术对成纤维细胞DNA水平、细胞周期分析。结果:实验组加入滇丹参后,细胞生长密度降低,梭状突起回缩,细胞双极变得圆顿,细胞形态变得不规则。与阴性对照组相比,加入滇丹参组成纤维细胞的生长受到明显的抑制(P<0.05),较高质量浓度(0.4,0.2g/mL)滇丹参对瘢痕成纤维细胞生长和抑制比低质量浓度(0.1,0.05g/mL)和rhIFN-α2a1000U/mL组明显(P<0.05)。增殖期(S-G2-M)细胞百分数明显降低,细胞凋亡水平明显提高。结论:滇丹参对瘢痕成纤维细胞的生长、增殖及胶原合成有抑制作用,不同的质量浓度作用效果有差别。BACKGROUND: It has proved that Salvia mittiorrhiza exhibits an important role in the process of wound healing. Salvia yunnanensis C.H.Wright has basically identical chemical composition, especially, superior in certain active component to Salvia miltiorrhiza. OBJECTIVE: To observe the effect of Salvia yunnanensis C.H.Wright on scar-derived fibroblasts, further more, to explore its function and mechanism to scars. DESIGE, TIME AND SETTING: The group control observation was performed in Kunming Medical College between March 2005 and March 2006. PARTICIPANTS: The scars were harvested from 8 patients, including 5 male and 3 female, with aged 7-38 years. The pathological course of scar was 1-12 years. Informed consent was obtained from each patient. METHODS: The scar-derived fibroblasts were randomly divided into the experimental, negative control, and positive control groups, following cultured in solution containing different concentration Salvia yunnanensis C.H.Wright (0.1 g/mL, 0.2 g/mL, 0.4 g/mL, 0.6 g/mL) for 24 hours in vitro, DMEM culture medium and DMEM culture medium with 1 000 U/mL rhlFN-a2a were added, respectively. MAIN OUTCOME MEASURES: DNA level, as well as cell cycle, was analyzed by light microscope, MTT assay, and flow cytometry, respectively. RESULTS: After adding with Salvia yunnanensis C.H.Wright, the density of cells depressed and the spindle-shape cell were discovered becoming retractation and abnormity. Compared with the negative control group, cell growth was significantly inhibited in the experimental group (P 〈 0.05). Salvia yunnanensis C.H.Wright with high concentration (0.4, 0.2 g/mL) had obvious inhibition on cell growth than that of the low concentration (0.1,0.05 g/mL)and positive control groups (P〈 0.05). The percentage of the cells in the growth phases (S-G2-M) of cell cycle decreased and the level of apoptosis increased in the experimental groups.
