通过体外受精技术缩短绒山羊育种周期的研究

摘　　要：以羔羊卵巢卵母细胞为材料，把体外受精技术应用到绒山羊育种工作中，观察了羔羊体外受精卵的体外发育及移植产羔的效果，以便探讨通过体外受精技术缩短绒山羊育种周期的技术途径．对４０６枚羔羊体外受精卵用Ｍ１９９＋ｈｃＧ、Ｍ１９９＋ＬＨ和Ｍ１９９＋ｈｃＧ＋ＥＧＦ三种培养基进行体外发育培养．结果：２～４细胞期胚的发育率平均分别为４０．０％、４３．６％和４９．６％；桑椹胚和囊胚的发育率平均分别为１７．５％、１５．８％和１８．８％．将４９枚Ｆ１代羔羊２～８细胞期胚移植给３１只受体母羊，结果有１１只妊娠（３５．５％）并产羔１２只（Ｆ２代）；将２４枚Ｆ２代羔羊２～４细胞期胚移植给１５只受体母羊，结果有２只妊娠（１３． The kid′s oocytes were fertilized in vitro,and the in vitro development of IVF embryos and the kidding after embryo transfer were observed.The purpose is to establish a method for shortening the breeding duration of cashmere goat.Four hundred and six kid′s fertilized oocytes were cultured in M199+hcG,M199+LH and M199+hcG+EGF respectively,the percentage of 2～4 cell stage embryos were 40.0%,43.6% and 49.6%,the percentage of morule and blastocysts were 17.5%、15.8% and 18.8%,respectively.Forty nine 2～8 cell stage embryos obtained from F1kids were transferred to 3 1recipients,the pregnancy rate was35.5%(11/31)and 12 F2 kids were delivered.Twenty four 2～4 cell stage embryos abtained from F2 kids were transferred to 15 recipients,two recipients became pregnant(13.3%)and 2 F3 kids were delivered.
