摘  要:目的:研究辛伐他汀诱导对骨形成蛋白-7(BMP-7)基因修饰的间充质干细胞(MSCs)增殖和分化表型的影响。方法:将辛伐他汀诱导和未经诱导的兔MSCs分别转导BMP-7基因后,Westernblotting检测转导后细胞BMP-7的表达;通过观察细胞生长以及碱性磷酸酶活性和测定骨钙素含量,分析辛伐他汀诱导对BMP-7基因修饰的MSCs的增殖和分化表型的影响。结果:转导后BMP-7基因在诱导组和未诱导组MSCs中均有表达,诱导组细胞BMP-7表达量、碱性磷酸酶活性和骨钙素含量均比较未诱导组明显增高。结论:转导前辛伐他汀诱导能够促进BMP-7基因修饰的MSCs的成骨转化。Objective To determine whether or not the induction of simvastatin influences proliferation and differentiation of the phenotype of bone morphogenie protein-7 (BMP-7) modified rabbit mesenehymal stem cells (rMSCs). Methods The rMSCs treated and untreated with simvastatin were transfeeted with BMP-7 gene. The expression of BMP-7 was determined by Western blotting. The changes of proliferation and differentiation of the phenotype in the cells were observed by cell growth state, alkaline phosphatase (ALP) activity and osteoealein (OCN) content analysis. Results The expression of BMP-7 was confirmed in levels of protein. Cell cultures of rMSCs pretreated with simvastatin showed a higher BMP-7 protein expression, ALP activity and osteoealcin content than that of untreated cells. Conclusion Early osteoblastic differentiation induced by Simvastatin can significantly promote BMP-7 modified MSCs osteohlast conversion.
