摘 要:目的:探索一种异源杂交瘤细胞株建立的方法.方法:以CSFV分点肌肉注射健康猪,成功诱导机体产生抗CS-FV的抗体.取免疫猪脾脏与鼠骨髓瘤细胞NS0融合,经筛选与克隆以获得猪源抗CSFV的mAb,并测定mAb的中和性及其效价.结果:成功筛选到15株抗CSFV的猪源mAb,中和实验结果表明9株mAb具有中和性,且同样识别重组的E2蛋白.结论:猪源mAb的成功制备为揭示CSFV在猪体内的免疫机制奠定了基础.AIM: To search for a method to prepare a swinemouse hybridoma. METHODS: Healthy swines were immunized with classical swine fever virus (CSFV) by intramuscular injection to induce anti-CSFV antibody. B cells of the immunized swine were fused with mouse myeloma cells NS0 and swine-mouse hybridoma was selected by CSFV and E2 protein. Neutralization test was carried out to identify the neutralizing titers of the swine antibodies. RESULTS : Fifteen strains of anti-CSFV swine-mouse monoclonal antibodies (mAb) were obtained after several times of screening and cloning, 9 of which were neutralizing antibodies and were recognized by recombinant E2 protein. CONCLUSION: The swine mAb may provide a valuable basis for further research of immunologic mechanism of CSFV in swines.
