等位基因特异性聚合酶链反应检验多药耐药基因的多态性

摘  要:目的:建立分析MDR1基因-3435C/T多态性等位基因特异聚合酶链反应(AS-PCR)的方法,并分析中国汉族人群中这个位点的基因型和等位基因频率。方法:对202例中国汉族健康男性采用等位基因特异聚合酶链反应和核苷酸序列测定技术分析其MDR1基因-3435C/T多态性的基因型和等位基因频率。结果:MDR1基因-3435多态性位点有CC、CT、TT3种基因型,频率分别为32.2%,54.4%,13.4%。C和T等位基因频率为59.4%和40.6%。结论:等位基因特异聚合酶链反应(AS-PCR)简便、准确、快速,适合于MDR1-C/T基因多态性检测。OBJECTIVE To establish the method of Allele-specific Polymerase Chain Reaction for detection MDR1 gene- 3435C/T polymorphisms in the 26 exon region of the MDR1 gene and investigate the polymorphisms in Chinese. METHODS Allele-specific Potymerase Chain Reaction and nucleoside sequencing were used to analyze the allelic frequencies and the genotype frequencies of MDR1 gene-3435C/T polymorphisms in two hundreds and two Chinese. RESULTS three genotypes of CC, CT,TT were identified in 3435 polymorphisms, the genotype frequencies of CC, CT,TT were 32. 18%, 54. 45 %, 13. 37%, respectively. While the allelic frequencies C and T were 59. 40% and 40. 60% respectively. CONCLUSION the Allele-specific Polymerase Chain Reaction is a simple, specific, and reliable method to detect the MDR1 gene-3435C/T polymorphisms in the 26 exon region of the MDR1 gene.
