摘  要:目的:优化黄芪总皂苷的纯化工艺。方法:以总皂苷含量为指标,采用均匀设计法对黄芪总皂苷的纯化工艺参数进行优化。结果:最佳纯化工艺条件为:药液浓度为1g原生药/ml,加入氢氧化钠至5%,加入水饱和正丁醇动态萃取2次,每次1.25倍量,加5%氢氧化钠溶液1倍量动态洗涤1次,加水1倍量动态洗涤2次,调节正丁醇溶液pH至中性。优化条件下的验证试验得到的黄芪总皂苷含量平均值为65.23%(n=3)。结论:建立的黄芪总皂苷的纯化工艺稳定、可行,适合放大生产。Objective: To investigate the optimum parameters to purify total saponin from Radix Astragali. Methods: the content of total saponin was set as the target.The uniform design method was applied to investigate the optimization process of purification. Results: The optimum condition of purifying the total saponin was obtained: the extract solution concentration of Radix Astragaliwas set at 1 g/ml; water saturation butanol was added to dynamic extract for two times with one and one-quartertimes of the volume each time; five percent of sodium hydroxide solution was added and given dynamic washing for one time; the water was used for dynamic washing for two times;then the butanol solution was regulated to neutral pH.The average content of total saponin was 65.23 % at the optimum purificaton condition in verification experimentations(n=3). Conclusions: The optimum parameters to purify the total saponin from Radix Astragali showed good stabilization and feasibility,and can be available for industrial production at large scale.
