HPLC法同时测定陈皮饮片中5种黄酮类化合物的含量

摘　　要：目的:建立高效液相色谱法同时测定陈皮药材中5种黄酮类化合物柚皮芸香苷、橙皮苷、川陈皮素、3,5,6,7,8,3’,4’-七甲氧基黄酮及红橘素的含量,并对结果进行聚类分析。方法:采用Waters Symmetry C18色谱柱(250mm×4.6mm,5μm);流动相为0.1%甲酸水溶液-乙腈,梯度洗脱;检测波长为300nm;流速1.0mL.min-1。结果:柚皮芸香苷、橙皮苷、川陈皮素、3,5,6,7,8,3’,4’-七甲氧基黄酮及红橘素的线性范围分别为1.00～9.00μg.mL-1(r=0.9998)、1.00～9.00μg.mL-1(r=0.9999)、0.10～0.90μg.mL-1(r=0.9997)、0.10～0.90μg.mL-1(r=0.9999)、0.10～0.90μg.mL-1(r=0.9994);加样回收率(n=5)分别为96.8%,99.4%,93.3%,94.4%,95.8%。结论:该方法准确、简便,具有良好的重复性和稳定性。试验结果显示,全国不同省市、不同产地陈皮饮片的含量存在差异。同时也说明多指标成分定量分析比单一指标成分定量分析能更全面地反映陈皮饮片的内在质量。Objective ： To develop an HPLC method for determination of contents of five flavonoids （ narirutin, hesperidin, nobiletin, 3,5,6,7,8,3＇, 4＇ - heptanmethphoxyflavone, tangeretin ） in Pericarpium Citri Reticulatae, and to accomplish the cluster analysis of contents. Method：HPLC was performed on a Waters Symmetry C18 analytical column（250 mm ×4.6 mm,5 μm） with gradient elution of 0. 1% aqueous formic acid and acetonitrile at the rate of 1.0 μg·mL^-1. The diode array detection was set at 300 nm. Results： Narirutin, hesperidin, nobiletin,3,5,6,7,8, 3＇,4＇ - heptanmethphoxyflavone and tangeretin had good linearity in the ranges of 1.00 - 9.00 μg·mL^-1 （r = 0. 9998） ,1.00 -9.00μg·mL^-1 （r =0. 9999） ,0. 10 -0. 90 μg·mL^-1 （r =0. 9997） ,0. 10 -0. 90 μg·mL^-1（r = 0. 9999） and 0. 10 - 0. 90 μg·mL^-1 （ r = 0. 9994 ）, respectively ; The average recoveries （ n = 5 ） were 96. 8%, 99.4% ,93.3% ,94.4% and 95.8% , respectively. Conclusion： The developed method is rapid, accurate with high repeatability. The results showed that the significant difference in samples from different provinees had been observed. The results also showed multiple chemical markers provided more comprehensive information than unique marker for quantitative analyses of Pericarpium Citri Reticulatae.
