摘  要:目的研究增龄进程中大鼠淋巴细胞中核转录因子κB(NF-κB)信号通路跨膜受体的mRNA水平差异表达特征,以及淫羊藿总黄酮(EF)对其的干预效应。方法将SD大鼠随机分为3天、4月龄、10月龄、18月龄、27月龄、27月+EF,以大鼠脾脏分离的淋巴细胞为研究对象,定制NF-κB信号通路基因芯片,分别检测各组NF-κB信号通路上游跨膜受体mRNA水平的整体表达特征及差异,并观察EF对NF-κB相关基因的干预效应。结果Tgfbr2、Tlr7、Ripk2出生3天表达最低,4或10个月达峰值,之后渐低,至27个月降至最低,EF干预后Ripk2变化较明显,接近10个月水平;Tnfrsf1a与Tlr2在3天时达峰,4个月骤然降低,而后在10、18、27个月均维持在一相近的水平,但经EF作用后有显著升高;Tnfrsf5自3天至18个月逐渐升高,但至27个月骤然下降,EF作用后略有回升;Madh4表达比较特殊,在4个月最高,10个月最低,18个月显著升高,至27个月再次降低,EF干预后略有提高,但变化不明显;Tnfrsf10b、Il1r1、Tlr1、Tlr4在增龄进程中的变化不明显。结论淫羊藿总黄酮的提前干预可对增龄进程淋巴细胞NF-κB信号通路跨膜受体基因mRNA表达产生一定的良性影响,尤其是对Ripk2的作用最为显著,这可能是EF作用的重要靶点之一。Objective To study the lymphocyte NF-κB signal passage cross membrane acceptor mRNA expression characteristics of rats in increasing age and the intervention effect of total flavone of Epimedium (EF). Methods The SD rats were randomized into 6 groups: 3 days (3d), 4 months (4m), 10 months (10m), 18 months (18m), 27 months (27m), 27m+EF. The lymphocytes separated from the spleen of rats were taken as the object and the NF-κB signal passage gene chip was used to examine the expression characteristics and differences of NF-κB signal passage upstream cross membrane acceptor mRNA of each group and to observe intervention effect of EF on NF-κB related gene. Results The expression of Tgfbr2, Tlr7, Ripk2 of 3d was the lowest, reached the peak 4m or 10m, afterward gradually lowered, dropping to the lowest 27m, with EF intervention, Ripk2 changed obviously, close to 10m level; Tnfrsfla and Tlr2 reached the peak on the 3d, in the 4m reduced suddenly, then in 10m, 18m and 27m maintained at a similar level, but with the EF intervention they were obviously increased; Tnfrsf5 increased gradually from 3d to 18m, but dropped suddenly in 27m, with EF intervention, it was slightly increased; the Madh4 expression was quite special, to the highest in 4m, to the lowest in 10m, increased obviously in 18m, reduced again in 27m, with EF intervention, slightly increased; the change of TnFrsf10b, Illrl, Tlr1, Tlr4 in increasing age was not obvious. Conclusion In the senile advancement, lymphocyte NF-κB signal passage cross membrane aeceptor mRNA increasing age dependence expression regulation characteristics were not inconsistent, the EF intervention produces a certain good influence on the old age group for gene mRNA expression, particularly a remarkable effect on Ripk2. This is possibly one of important targets of EF functions.
