犬瘟热病毒强毒株的分离与鉴定

摘　　要：从病死犬肺、肝和淋巴结分离到２株ＣＤＶ，ＣＤＶ９３０３９和ＣＤＶ９３０４１。消除小牛血清中的干扰因素和３３℃培养是分离成功的主要因素。两株病毒均在Ｖｅｒｏ细胞生长良好，传２～４代毒力稳定，ＣＰＥ产生明显，ＴＣＩＤ５０为１０－６．５０～１０－６．７７，明显高于国内外野毒株和弱毒株的毒力效价。人工感染犬死亡率较高，表明该分离株致病力较强，作者推测我国犬群可能存在ＣＤＶ强毒变异株。オwo　strains　of　canine　distemper　virus　(CDV)　were　directly　isolated　from　the　lung,liver　and　lymph　node　of　infected　dogs　with　vero　cell　cultures.Elimination　of　certain　interfering　factors　that　calf　serum　contains,and　incubation　at　33℃　were　the　main　factors　for　a　successful　isolation.The　two　strains　of　CDV　could　grow　very　well　in　vero　cell　cultures.The　growth　properties　were　cytopathogenic,inducing　cellular　necrosis,syncytium　formation,and　inclusion　bodies　within　infected　cultures.They　did　not　give　hemagglutination　with　human,mouse,rat,guinea　pig,rabbit,dog,monkey,chicken,pig,sheep　and　Chinese　hamster　red　blood　cells.Under　electron　microscope,the　capsid　was　enclosed　by　envelope　which　carries　an　outer　fringe　of　fine　spikes　or　projections.Two　strains　readily　proliferated　in　vero　cell　cultures　,the　mean　tissue　culture　infective　doses　(TCID50)were　10-6.50～106.77from　2　to　4　generations.That　was　higher　than　that　of　virulent　field　strains　and　vaccine　strains.The　puppies　individually　infected　with　two　strains　by　intraperitoneal　inoculation　and　intranasal　inoculation　showed　distinctive　clinical　signs,and　the　mortality　was　high.The　authors　suspect　that　virulent　variant　strains　of　CDV　may　exist　in　China.
