摘 要:[目的]研究急性酒精摄入对大鼠记忆功能的影响,及海马内cAMP反应元件结合蛋白(CREB)磷酸化水平的变化。[方法]采用Morris水迷宫方法检测急性摄入1g/kg、2g/kg和3g/kg剂量的酒精,以及摄入3g/kg酒精0.5、1、2、4h后成年雄性SD大鼠记忆功能的变化,采用Western Blot方法检测急性摄入3g/kg后大鼠海马中CREB磷酸化(p-CREB)水平随时间的变化。[结果]Morris水迷宫检测结果显示,随酒精摄入剂量增加,潜伏期及游泳路程均呈延长趋势,其中2g/kg组和3g/kg组与对照组比较差异有统计学意义(P﹤0.05);急性摄入3g/kg酒精0.5、1及2h后,潜伏期及游泳路程明显延长(P﹤0.05),其中2h最高。4h恢复。WesternBlot结果显示急性摄入3g/kg酒精后0.5、1及2hCREB磷酸化水平逐渐升高,2h达到最高,4h降至正常水平。[结论]急性摄入酒精引起记忆功能改变存在量效、时效规律,大鼠海马中CREB磷酸化水平变化与其相一致,提示酒精可能通过引起海马内记忆功能相关蛋白CREB磷酸化水平升高引起记忆功能的改变。[Objective] To observe the effect of acute alcohol administration on memory-impairment and phosphorylation level of cAMP-responsive element binding protein (CREB) of hippocampus in rats. [Methods] Alcohol was administrated to rats by intragastric lavage with doses of 1, 2 and 3g/kg. 2 hours later, Morris Water Maze performance was applied to evaluate memory function, and the phosphorylation ievel of cAMP-responsive element binding protein in rat hippocampus was determined by Western Blotting. Also, memory function and p-CREB level were determined at 0.5, 1, 2 and 4 hour after administration with alcohol 3g/kg. [ Results ] The Morris Water Maze performances demonstrated that when the alcohol doses increasing, latency and traveling distance prolonged, with significant differences between 2g/kg, 3g/kg dose groups and the control (P 〈 0.05) ; The latency and traveling distance extended 0.5, 1 and 2 h after administration with alcohol 3g/kg with significant difference in contrast to the control group (P 〈 0.05): The Western Blotting results indicated that the phosphorylation level of CREB elevated gradually after administration, waking at 2h and returning to normal content af 4h. [ Conclusion] The increase of phosphorylation level of CREB in rat hippoeampus is in accordance with the instance of memory-impairment after acute lavage with alcohol, which implies that alcohol may induce memory-impairment by enhancing the phosphorylafion level of CREB.
