摘  要:目的采用RP-HPLC法,测定兔血浆中的替普瑞酮。方法血浆样品中加入丁烯二酸二异辛酯和NaH2PO4溶液(0.05mol·L-1,pH2.5),用叔丁醚振荡萃取后测定。色谱柱为LunaC8柱(150mm×4.6mm,5μm),流动相为乙腈-水(92:8),流速1.0m.lmin-1,柱温25℃,检测波长205nm。结果替普瑞酮0.086~3.440μg.ml-1线性关系良好(r=0.9991),平均方法回收率为98.25%,日内和日间RSD均小于10%。结论所建方法简便、灵敏、准确、重复性好,适用于替普瑞酮的药物动力学研究。OBJECTIVE A simple RP - HPLC method was established for determination of Teprenone in rabbit plasma. METHODS Internal standard diisooctyl maleate and NaH2PO4 (0. 05 mol·L-1 ,pH2. 5)were added to plasma sample, then the sample was extracted with tertbutylmethylether and the extract was determined on a Cs analytical column( 150 mm× 4. 6 mm, 5 μm). The experi- ment was performed with mobile phase of aeetonitrile - water ( 92 : 8 ) at a flow rate of 1.0 ml·min-1and detected at 205 nm. RESULTS The linear range was 0.086 - 3. 440μg· ml- 1 ( r = 0. 9991 ). The analytical recovery was 98.25 %, and the RSD of within -day and between -day RSD was less than 10%. CONCLUSION This method is simple, sensitive, accurate and reproducible. It is applicable for pharmacokinetic study of Teprenone.
