复方丹参滴丸对大鼠动脉粥样硬化及TAFI水平的影响

摘　　要：目的通过建立大鼠动脉粥样硬化(AS)模型,探讨复方丹参滴丸通过调节凝血酶活化的纤溶抑制剂(thrombin-activatable fibrinolysis inhibitor,TAFI)干预AS的作用机制。方法40只健康雄性的Wistar大鼠随机分为正常对照组、非药物治疗组、高剂量药物治疗组、低剂量药物治疗组,每组10只。正常对照组给予普通饲料喂养,3个模型组给予高脂饲料喂养的基础上行主动脉球囊损伤术,喂养4个月,以复制大鼠AS模型。用全自动生化仪测定血脂系列,发色底物法测定血浆TAFI的活性。结果成模后,3组模型鼠的血浆总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和TAFI活性明显高于正常对照组(P<0.01);药物治疗后,与非药物治疗组相比,高剂量组与低剂量组的TC、TG、LDL-C和TAFI活性明显低于非药物治疗组(P<0.01),与低剂量组间比较,高剂量组TC、TG、LDL-C和TAFI活性明显低于低剂量组(P<0.05)。结论AS过程中TAFI水平升高,使用复方丹参滴丸干预后,TAFI水平下降,提示复方丹参滴丸可能通过下调TAFI水平进而干预了AS的形成。Objective To investigate the effect of Fufang danshen diwan on atherosclerosis （AS） by lhe mechanism of thrombin-activatable fibrinolysis inhibitor （TAF1） using AS rat. Methods 40 healthy male Wistar rats were randomly divided into control, non-drug treatment, Fufang danshen diwan high and low dose groups, 10 rats in eaeh group. Last three groups were model groups. Control group were fed with normal diet, and the other model groups were fed with high lipid and given aorta endothelium injury. In order to copy AS model rats, the four groups were fed four months. Then the levels of TC, TG, LDL-C and TAFI were measured by ehromogenic assay. Results The levels of TC, TG, LDL-C and TAF1 in three model groups were higher than those in control group（P 〈 0. 01 ）. After treatment, the levels of TC, TG, LDL-C and TAFI in Fufang danshen diwan group were lower than those in non-drug treatment group obviously（ P 〈 0. 01 ） , and the levels of TC, TG, LDL-C and TAFI in Fufang dansheu diwan high dose group were lower than those in Fufang danshen diwan low dose group （P 〈 0. 05）. Conclusions The increased level of TAFI is in the progress of AS could be decreased after treated with Fufang danshen diwan, which indicates Fufang danshen diwan interfere the process of AS through down regulating the level of TAFI.
