摘 要:目的研究拉伸刺激下力生长因子(mechano—growth factor,MGF)在成骨细胞中的表达和分布。方法对成骨细胞施加周期性动态拉伸刺激,并对MGF的表达做蛋白免疫印记和细胞免疫荧光分析。结果蛋白免疫印记分析显示,周期性拉伸刺激会使成骨细胞中MGF的表达量显著提高,加载12h对蛋白表达的促进作用最明显,蛋白质水平提高了4倍,随后急剧降低。细胞免疫荧光分析发现,MGF较为集中地分布在细胞核。结论拉伸刺激下,MGF在成骨细胞中出现一个短暂的表达高峰,这可能与拉伸所致的细胞损伤有关。MGF集中分布在细胞核,提示MGF含核定位信号,使得该因子参与其他基因的表达调控。Objective To analyze the protein expression and subcellular distribution of mechano- growth factor (MGF) in osteoblasts under stretch stimulation. Methods Cyclic stretching was applied to osteoblasts by a mechanical stretching device. The whole-cell proteins were extracted from controlled and stretched osteoblasts for detecting the protein expression level of MGF by Western blot and observing the intracellular distribution of MGF by fluorescent immunocytological method. Results Western blot showed significant increase of expression of MGF in osteoblasts under stimulation of cyclic stretching. The level of protein was increased by four folds after 12-hour stretching of osteoblasts, and then declined sharply. Immunofluorescence analysis showed that MGF was mainly distributed in the nuclei of osteo- blasts. ConclusionsUnder the cyclic stimulation, the expression of MGF reaches a short period of peak in osteoblasts, which may be related to the injury of osteoblasts caused by stretching. MGF is mainly distributed in the nuclei of osteoblasts, indicating that MGF may contain nuclear localization signal and modulate the expression of relative genes.
