摘 要:[目的]探讨大气PM2.5(Particulate Materials)水溶成分对大鼠离体胸主动脉收缩作用的影响及可能机制。[方法]Wistar雄性大鼠15只,分离胸主动脉制备内皮完整、去内皮血管环,分别观察0μg/ml、10μg/ml、100μg/mlPM2.5对不同浓度苯肾上腺素(PE)引发血管收缩的影响;内皮完整血管环同时以超氧化物歧化酶(SOD)联合二甲基亚砜(DMSO)、左旋硝基精氨酸甲酯(L-NAME)干预,探讨对100μg/mlPM2.5作用有无影响。[结果](1)内皮完整组:PE在10-5mol/L水平,对照组、10μg/ml组、100μg/ml组所引发收缩率递增分别为0.77±0.06、0.82±0.074、0.90±0.033(P﹤0.05);SOD联合DMSO可逆转100μg/mlPM2.5对收缩的影响。(2)去内皮组:PE各个浓度组在3组间收缩率均无差异。[结论]PM2.5可加强PE对血管环的收缩作用,其机制可能与其通过氧化应激致内皮细胞功能障碍有关。[Objective] To explore the effects of water-soluble components of Particulate Matter (PM2.5) in the air on the contraction in isolated rat thoracic aortas and its possible mechanisms. [ Methods]115 Wistar rats were isolated the thoracic aortas to prepare integrated endothelium and vascular ring Without endothelium, and the effects of 0 μg/ml, 10 μg/ml and 100 μg/ml PM2.5 on vasoconstriction induced by PE were observed. The effect of Phenylephrine (PE) with or without being pretreated with superoxide dismutase (SOD) combined with Dimethyl sulphoxide (DMSO) or only with NG -nitro- L-arginine methylester ester (L-NAME) on the contraction of intact endothelium rings were also observed. [ Results ] (1) On the level of 10^-5 mol/l for PE, The shrinkage rate due to the PM2.5 of 100μg/ml significantly higher than controls, and 10 μg/ml group was significantly higher than the 100 μg/ml group (P 〈 0.05 ), respectively. SOD combined with DMSO could reversed the effect of 100 μg/ml on contraction; (2) The rates of contraction did not show significantly different among the group without endothelium, and the three groups with different concentrations of PE. [Conclusion] PM2.5 can promote the contraction of PE in aortas, which possibly with the probable mechanism of oxidative impairment in endothelial cells.
