摘 要:目的:观察大鼠脂肪干细胞与2种不同去细胞神经支架的体外相容性,为周围神经组织工程寻求合适的种子细胞载体。方法:将大鼠脂肪干细胞与传统去细胞神经支架和改良法去细胞神经支架体外复合培养,采用倒置显微镜和扫描电镜观察细胞在2种支架上的生长、黏附情况,测定细胞活性及细胞吸附率,并与空白对照组比较。结果:脂肪干细胞可在2种去细胞神经支架上生长、黏附,在改良法去细胞神经支架上的细胞吸附率优于传统去细胞神经支架。结论:脂肪干细胞与2种去细胞神经支架均有较好的体外组织相容性,改良法去细胞神经支架可作为更有效的周围神经组织工程细胞载体。Aim: To study the compatibility between adipose derived stem cells and two different kind of acellular nerve scaffold in vitro in order to explore appropriate scaffold material for peripheral nerve tissue engineering. Methods: Adipose derived stem cells and two different kinds of acellular nerve scaffold were cocuhured in vitro,the growth and attachment of cells in scaffold were studied by inverted phase contrast microscope, the activity and adherence rate of cells were compared to the control group. Results: Adipose derived stem cells could grow and attach well in acellular nerve scaffold, the adherence rate of cells in scaffold of improved method were better than that of traditional method. Conclusion: There are better compatibility between adipose derived stem ceils and two kind of acellular nerve scaffold in vitro, the acellular nerve scaffold of improved method can be used as a better vehicle for seed cell of peripheral nerve tissue engineering.
