摘 要:【目的】对小麦类脱水素基因进行克隆与分析,为深入研究植物脱水素的结构及其在耐旱中的可能功能提供理论依据。【方法】利用PEG胁迫处理小麦幼苗,采用RACE方法,从干旱诱导的郑引1号小麦中获得脱水素基因,采用生物信息学方法对该基因进行耐旱功能分析。【结果】获得了小麦脱水素基因新成员WZY2的全长序列,其长度为849 bp,含1个459 bp的开放阅读框,编码152个氨基酸。该氨基酸序列具有明显的脱水素类蛋白特征(K片段),由2个K片段、1个S片段和1个Y片段组成,属于YSK2型脱水素。该脱水素具有较高的亲水性,表明该蛋白在束缚自由水、稳定膜结构方面有一定作用。【结论】从水分胁迫的郑引1号小麦中克隆的脱水素基因WZY2属于YSK2型脱水素。[Objective] The objective of this study was to isolate and clone the dehydrin gene from Zhengyin No. 1. [Method] Using the method of RACE, with PEG stress wheat seedlings, dehydrin gene from Zhengyin No. 1 was cloned and the sequence was analyzed by bioinformatics software. [Result] A full-length sequence of a new dehydrin gene was amplified;the gene has a length of 849 bp,ineluding a 459 bp ORF (open reading frame). Analysis shows that the protein sequence of the gene encoded has the characteristics of dehydrin genes:the K fragment, S and Y fragment. It is a YSK2 dehydrin formed by two K fragments,a S and a Y fragment. The high hydrophilicity of the protein indicates that it plays an important role in binding the free water and stabilizing the membrane. [Conclusion] The dehydrin gene WZY2 cloned from Zhengyin No. 1 provides further information on the structure of dehydrin and it has possible function in plant tolerance to drought.
