紫穗槐核型分析方法的探讨

摘　　要：采用低温0.1％秋水仙素、0.002mol／L 8-羟基喹啉溶液，3种预处理方法对紫穗槐的根尖进行处理。结果表明：以0-4℃处理36h效果最佳。紫穗槐的体细胞染色体数目为2n=40，核型公式K（2n）=2x=40=28m＋8sm＋2st＋2m（SAT），核型分类为“2B”型，不对称系数57.70％，为较对称类型。Using low temperature, 0.1% colchicine treatment, 0.002 mol/L 8-hydroxy quinoline solution processing, three methods to treatment the root tip of Amorpha fruticosa. The results showed that using low temperature with 36 hours was the best way. The number of chromosomes of Amorpha fruticosa was 2n=40, the karyotype formula was K（2n）= 2x=40=28m＋8sm＋2st＋2m（SAT）, belonging to “2B” type. The comfficient of non-symmetry was 57.70 %, was a more symmetrical type.
