摘  要:目的探索大鼠骨髓源性肝干细胞(BDHSC)脾内移植后的分化情况。方法利用肝细胞生长因子(HGF)体外诱导大鼠BDHSC,并植入60只成年SD大鼠脾内,两组(n=30)分别于术后7天和14天处死动物后取脾,RT-PCR及免疫组化检测移植细胞的AFP及白蛋白的表达。结果移植后7天,AFPmRNA、白蛋白mRNA和AFP的阳性表达率分别为73.3%、46.7%、56.7%;移植后14天,AFPmRNA阳性表达率(23.3%)显著下降(P<0.01),白蛋白mRNA阳性表达率(20%)显著下降(P<0.05),AFP阳性表达率(26.7%)亦显著下降(P<0.05)。结论体外诱导大鼠BDHSC脾内移植后。Objective To explore the differentiation of rat bone marrow derived hepatic stem cell (BDHSC) after intrasplenical transplantation. Methods BDHSC was induced by hepatocytes growth factor in vitro, 7 days and 14 days after intrasplenical transplantation, 60 rats(two groups, n=30) were separately put to death for spleen, RT-PCR and immunohistochemistry were used to examine the expression of AFP and albumin in the BDHSC. Results 7 days after transplantation, the positive rate of AFP mRNA,albumin mRNA and AFP was 73.3% ,46.7% ,56.7%; 14 days after transplantation, the positive rate of AFP mRNA (23.3%) decreased significantly(P〈0.01), the positive rate of albumin mRNA(20% ) decreased significantly(P〈0.05), and the positive rate of AFP(26.7%) also decreased significantly(P〈0.05). Conclusion BDHSCs in vitro might gradually lose their acquired characters of hepatocytes after intrasplenical transplantation.
