胶质细胞参与电针足三里对胃运动的调节作用

摘　　要：目的观察迷走复合体(dorsal vagal complex,DVC)中神经元和胶质细胞的形态变化,并采用胶质细胞代谢阻断剂丙戊茶碱(propentofylline,PPF)予以干预,研究胶质细胞是否参与针刺足三里对胃运动功能调节作用的影响,探讨针刺对胃运动调节作用的神经机制。方法采用免疫荧光组织化学方法,观察大鼠中枢核团DVC中神经元被激活的标志c-fos以及星形胶质细胞GFAP、小胶质细胞OX42的表达变化。采用PPF予以干预,观察上述指标的变化。并同步测定大鼠胃电活动的变化。结果对照组大鼠DVC的c-fos呈现低水平表达,GFAP和OX42活化程度很低,胃电亦较为规律。电针足三里组大鼠DVC的c-fos表达水平、GFAP和OX42活化程度明显增加,胃电的波幅和频率均有显著增加。丙戊茶碱干预后电针足三里组大鼠DVC的c-fos表达水平、GFAP和OX42活化程度明显降低,胃电的波幅和频率也有显著的降低,与无干预组相比差异有显著性。但与对照组相比差异依然显著。结论电针足三里对胃运动功能的调节不但有神经元的作用,而且胶质细胞也参与这种调节作用。[Objective] To observe the changes of neurons and glial cells （astrocytes and microglial） in morphologic in dorsal vagal complex （DVC） after electro-acupunctured （EA） at Tsusanli, then by using the propentofylline （PPF） blocked the metabolic function of glials, to investigate the glials possibly participate in electro-acupuncture at Tsusanli regulated gastric activity. [Methods] By using the immunofluorescent staining method and electrophysiological method, observed the expressions of c-fos GFAP and OX42 of DVC after EA at Tsusanli in rats under using or un-using the metabolic blocking-agent via an injection in the cavitas subarachnoidealis. In the mean time, the EGG was measured synchronous in rats. [Results] The expressions of c-fos GFAP and ox42 in DVC were low level in control group, The EGG was discipline. But the expressions of c-fos GFAP and OX42 were obvious in DVC after EA at Tsusanli compared with control group. The amplitude and frequency of EGG were different from the control group. The level of expressions of c-fos GFAP and OX42 in DVC decreased compared with control group and EA at Tsusanli group after using the propentofylline then EA at Tsusanli. But in the group of an injection in the cavitas subarachnoidealis with identical volume normal sodium, the level of expressions of c-fos GFAP and 0X42 in DVC have no obvious changes compared with EA at Tsusanli group. The changes of EGG were similar to the expressions of c-fos GFAP and OX42. [Conclusion] （;has of the dorsal vagal complex possibly participate in electro-acupuncture of Tsusanli regulated gastric activity.
