摘 要:目的诱导获得人外周血巨噬细胞(MΦ),研究嗜酸乳杆菌作用MΦ后对其抗肿瘤效应的影响及机制。方法自正常人外周血分离获得单核细胞,体外rhGM-CSF诱导培养,观察细胞形态并检测其相关表型;利用MTT法检测所诱导MΦ对宫颈癌细胞HeLa的直接抑瘤效应;ELISA法检测其培养上清中TNF-α及IL-12的含量。结果诱导7d的贴壁细胞具有典型的MΦ形态,流式分析显示为分化成熟的MΦ;体外细胞毒实验显示:嗜酸乳杆菌活菌状态和热灭活状态以10∶1、1∶1与MΦ相互作用后能够显著增强MΦ对HeLa细胞的杀伤作用(P<0.05),激活后的MΦ在一定效靶比为10∶1时其肿瘤抑制率最大,与LPS阳性对照相比,其抗瘤活性差异不存在显著性(P>0.05);经嗜酸乳杆菌活菌和热灭活菌作用的MΦ能显著提高IL-12和TNF-α两种细胞因子的分泌量,而且存在剂量依赖性,与空白对照组比较差异存在显著性(P<0.05)。结论两种生物状态的嗜酸乳杆菌均能够增强人MΦ抗宫颈癌细胞作用。[Objective] To induce macmphages (MФ) from human peripheral blood in vitro, and to detect the anitimor effect of MФ mediated by lactobacilli, further to explore the mechanism. [Methods] The monocytes were acquired from the human peripheral blood and were cultured with rhGM-CSF. The mophology of non-adherent cells was observed by microscope and the related phenotypes of the cells were detected by Flow cytometry. MTT was used to detect the direct antitumor effect of MФ to cervical cancer cell HeLa. ELISA was used to detect the quantity of TNF-α and IL-12 in supernatant of MФ. [Results] After 7 days'culture, the non-adherent cells had the typical morphology of MФ. Flow ctometry analysis showed theses cells were defferentiated and ripe MФ. Living and heatkilled lactobacilli could significantly increase the antitumor effect of MФ to HeLa when interacted with MФ at ratio of 10:1, 1:1 (P 〈0.05), activated MФ had the best tumor control rate when the ratio of effect and target is 10:1, its antitumor effect had no significant difference compared with positive control LPS (P 〉0.05). MФ had more secretory volume of IL-12 and TNF-α activated by two forms of lactobacilli with dose dependent, compared with control (P 〈 0.05). [ Conclusions] Two forms of lactobacilli could increase antitumor effect of human MФ.
