摘 要:大麦糖信号转录因子SUSIBA2是WRKY超基因家族的一员,通过识别某些淀粉合成酶基因启动子区的糖应答元件调控胚乳中淀粉的积累.本研究根据SUSIBA2序列,从水稻基因组中搜索到1个与SUSIBA2相似的基因,称为SUSIRI.利用PCR方法,从水稻基因组DNA中克隆出该基因第3外显子455 bp的片段,它包含1个WRKY保守结构域.以此作为干扰序列,构建了干扰载体.通过农杆菌介导法将其转化水稻,获得了16株T0代阳性植株.SUSIBA2, a transcription factor for sugar signaling in barley, belongs to the WRKY super gene family. It regulates starch accumulation in endosperul by recognizing sugar-responslve elements in the promoter regions of genes for starch synthesis. According to the SUS[BA2 sequence, we identified a SUSIBA2-1ike gene ( named SUSIRI) by blasting the rice genome in GeneBank. A frag- ment in the third exon of this gene was cloned by PCR from rice genomic DNA, which was 455 bp in length and contained a conser- vative WRKY domain. An RNAi vector was constructed using the cloned fragment as the interference sequence. The RNAi fragment was then transferred into recipient genome of rice mediated by Agrobacterium tumefacieas and 16 positive To plandets were obtained.
