摘  要:应用高效液相色谱法建立欧洲鳗鲡血浆和肌肉中甲苯咪唑(MBZ)及其代谢物5-羟甲基苯基苯并咪唑(MBZ-OH)和二氨基-5-苯基苯并咪唑(MBZ-NH2)浓度的检测方法.结果表明:选用C18色谱柱,检测波长为248 nm,样品经Na2CO3溶液和乙酸乙酯提取,以乙腈和醋酸—醋酸钠缓冲液(pH 4.5)为流动相,在0.01-1.00μg.mL-1内,MBZ及其代谢物的线性关系良好,标准品添加平均回收率均大于90%,日内、日间变异系数均低于6.70%,最低检测限为3 ng.mL-1.A high-performance liquid chromatographic method for the determination of mebendazole (MBZ) and its metabolites hydroxy-mebendazole (MBZ-OH) and amino-mebendazole (MBZ-NH2 ) in European eels (Anguilla anguilla) plasma and muscle tissue was developed. The results showed that under the condition of extracting plasma and muscle tissue with Na2CO3 solution and ethyl acetate, analyzing on a C18 column, taking acetouitrile and acetate acid-sodium acetate buffer (pH 4.5) as mobile phase, a linear range of applicability of 0.01 -1.00 μg·mL-1 was found for mebendazole and its metabolites. Mean recoveries were both over 90%, the coefficients of variation were less than 6.70% and the lowest detective limit was 3 ng ·mL-1.
