摘  要:为研究马传染性贫血病毒病(EIAV)减毒疫苗免疫动物后体液免疫反应的成熟时相,选用健康马4匹,2匹接种EIAV感染性克隆减毒疫苗pLGFD3毒后,另2匹接种生理盐水做对照,接种后定期采集血样,采用ELISA方法检测各个时相点血清中gp90,p26的结合抗体滴度,并采用细胞蚀斑染色检测病毒中和抗体滴度,分析抗体反应的规律。结果显示,感染性克隆pLGFD3毒免疫后,针对envgp90和gagp26蛋白均产生了特异性的体液免疫反应,gp90结合抗体在2个半月左右达峰值,随后尽管有轻微的波动,一直维持在较高的水平直至免疫后5~6个月,随后开始轻微回落,在免疫后32个月时仍能检测到抗体的稳定存在,而p26结合抗体在1~2个月左右达峰值后则呈明显回落,在4个月时降至检测水平以下。gp90诱导的特异性中和抗体的水平同样在2个月左右达峰值,在整个观测期内持续稳定存在。结果表明,EIAV感染性克隆减毒疫苗免疫后,能诱导产生特异性的gp90,p26结合抗体以及中和抗体,而且gp90特异性抗体和中和抗体可持续稳定存在,这些抗体可能参与减毒疫苗诱导的免疫保护作用。To investigate the kinetics of humoral immune response to EIAV vaccine, two horses were immunized with pLGFD3-v infectious clone derived from an attenuated by subcutaneous injection and blood samples were collected at various time-points for antibody detection during subsequent two and half years. EIAV infectious clone pLGFD3-v was shown to induce antibodies against both EIAV env gp90 and gag p26. The gp90-specific antibody level increased significantly and reached the plateau at two and half months, and remained detectable even at 32 months post inoculation. The neutralizing antibodies displayed a comparable kinetics. In contrast, the gag-specific antibodies peaked at 1 to 2 months and declined quickly afterwards and almost reached undetectable level within 4 months. These results demonstrated that pLGFD3-v could induce antibodies against both EIAV env gp90 and gag p26, and neutralizing antibodies. The prolonged lasting of env-specific antibodies and neutralizing antibodies may contribute to the immune protection conferred by EIAV attenuated vaccine.
