摘 要:目的评价谷胱甘肽(GSH)对顺铂(C-DDP)引起的肾毒性的影响。方法采用随机分组法对30只Wistar大鼠分成对照组、C-DDP组和C-DDP+GSH组。对照组给予生理盐水(20ml/kg,腹腔注射,每周2次,共5周),C-DDP组大鼠给予C-DDP(2mg/kg,腹腔注射,每周2次,共5周);C-DDP+GSH组除与C-DDP组给药相同外,另同时给予GSH(0.3ml/100g,腹腔注射)。5周后将大鼠处死。取肾脏进行病理检查及用电感耦合等离子体原子发射光谱(ICP-AES)法测定肾脏中铂的含量。结果肾组织中铂浓度结果显示,C-DDP组与C-DDP+GSH组铂浓度的差异没有统计学意义,说明GSH不影响C-DDP在肾脏中的蓄积量。肾脏病理组织学结果表明,C-DDP可引起肾脏组织学改变,而GSH可减轻C-DDP所致的肾损伤,说明GSH对C-DDP引起的肾损伤在病理上有一定程度的缓解。结论GSH在不影响C-DDP蓄积量的情况下,一定程度上可以中和C-DDP引起的肾毒性。[ Objective] To evaluate the effect GSH on nephrotoxicity induced by Cisplatin (C-DDP). [ Methods ] Wistar rats were randomly divided into three groups: control group, C-DDP group and C-DDP + GSH group. The control group was administered physiological saline { 20 ml/kg, intraperitoneal injection, 2 times per week for 5 weeks }. The C-DDP group was administered with C-DDP (2mg/kg, intraperitoneal injection, 2 times per week for 5 weeks). The GSH + C-DDP group was administrated with GSH {0.3 ml/100 g, intraperitoneal injection) at the same time in addition to the same treatment as C-DDP group. Rats were sacrificed after 5 weeks. The kidneys were performed by pathologic examination and the ptatinic content was measured by the method of ICP AES. [ Results] The concentration of platinum in kidney tissues showed that the difference of concentration of platinum between C- DDP group and C-DDP + GSH group was not significant, and indicated that GSH had no effect on the accumulation of C-DDP in kidney. Rerial pathology results showed that C-DDP can cause histological change of kidney but GSH can reduce the C-DDP-induced renal injury, and indicated that C-DDP can pathologically ease the GSH-induced kidney damage to some degree. [ Conclusion] Without affecting the accumulation of C-DDP, GSH can neutralize nephrotoxicity induced by C-DDP to a certain extent.
