库存软骨异体移植后的图像定量分析

摘　　要：０．４％戊二醛甲醛混合液保存４周的猪透明肋软骨异体皮下移植１～５月后图像定量分析移植软骨的细胞学形态参数及组织中ＧＡＧ（酸性粘多糖）的含量．结果表明：各实验组细胞密度均高于对照组（ｐ＜０．００１），证明软骨细胞继续保持其增殖分裂活性．保存未移植软骨，移植１月及３月后细胞形态因子、核周长、核面积ＤＮＡ相对含量等４项参数均无显著性差异（ｐ＞０．０５），而移植５月后该４项参数均与对照组有显著差异（ｐ＜０．００１），均不同程度下降达原对照组的８７．８％，８９．４％，７７．０％及７６．８％．核浆比值在保存及异体移植１～３月后升高，移植５月后仍继续升高达对照组的１．３１倍．ＡＢ染色切片细胞内的酸性粘多糖含量于移植１～５月内升高超过对照组含量，于５月末达到对照组的１０８．３％（ｐ＜０．００１），而基质中的ＧＡＧ含量在移植后１～３月时与对照组无明显差异（ｐ＞０．０５）而移植５月后下降（ｐ＜０．００１）但仍达对照组９９．７％．The present paper studied the cell's morphological parameters and the quantities of GAG by image analysis technigue in porcine cartilage histology which were preserved in 0.4% glutaraldehyde and formaldehyde mixture in a duration of 4 weeks,then allografted subdermaly during 1～5 mohths.The result showed:the cell's density of the various experiment groups were all higher than those of the control's(p<0.001).The result suggested that the cartilage cells kept the properity of mitosis and fertility. The 4 parameters about cell's morphological factor,nucler area and DNA reletative quantity among the preserving groups,groups of grafting after 1 month and 3 months were all with no significant difference.(p<0.05).But the 4 parameters of the groups of grafting after 5 months were with significant difference,compared with those of the normal control's(p<0.001),it reduced down to be 87.8%,89.4,77.0% and 76.8% that of the normal controls respectively.
