摘 要:目的探讨单次惊厥发作后脑内神经性钙黏附分子(N-Cadherin)表达与海马结构中苔藓纤维发芽现象之间的关系及其作用。方法利用腹腔注射戊四氮(PTZ)制作发育期SD大鼠(14天、28天)单次惊厥发作模型,各日龄组随机分为生理盐水(NS)组、PTZ致惊组和吡咯二硫氨基甲酸酯(PDTC)预处理组,采用免疫组化法检测N-Cadherin的表达,利用Timm染色法观察海马苔藓纤维发芽现象。结果NS组在海马CA3区可见极少Timm染色颗粒,致惊后1周偶见Timm染色颗粒,与NS组相比差异无显著性(P>0.05);致惊后3周可见较多Timm染色颗粒沿海马CA3区呈条带样分布,较NS组明显增加(P<0.01);PDTC预处理组可见Timm染色颗粒较PTZ致惊组明显减少(P<0.05)。同时,NS组14天和28天大鼠海马CA3区可见少量的N-Cadherin阳性细胞,着色不深;PTZ致惊组海马CA3和齿状回门区的N-Cadherin阳性细胞与NS组相比明显增多(P<0.01),PDTC预处理后相同区域内N-Cadherin阳性细胞较PTZ致惊组明显减少(P<0.05)。N-Cadherin与Timm染色颗粒的表达结果与惊厥鼠的日龄并无关联性。结论单次惊厥发作在幼鼠可以引起海马不同程度的苔藓纤维发芽,而N-Cadherin的分布位置与变化时相与苔藓纤维发芽相吻合,这表明N-Cadherin可能参与或促进了苔藓纤维的发芽。Objective To investigate the relationship between the expression of N - Cadherin in the brain and mossy fiber sprouting in the rat hippoeampus following single seizure in immature rats so as to ascertain the possible effect of N - Cadherin. Methods 96 immature rats of 14 d and 28 d each were induced single seizure by intraperitoneal injection of PTZ 60 mg/kg to establish animal model. The rats were randomly classified into three groups, including normal saline control (NS group), PTZ seizure group, pyrrolidine dithiocarbamate (PDTC) pretreatment group. Immunohistochemistry method was used to determine the expression of N - Cadherin in rat hippocampus after seizure, and Timm histochemical technique was adopted to observe mossy fiber sprouting. Results In NS group, Timm - stained granules were rare in the CA3 area of hippocampus, and in PTZ seizure group only a few Timm - stained granules were visible 1 week later. The difference between the two groups was of no statistic significance ( P 〉 0.05 ). Three weeks later, in PTZ seizure group, Timm -stained granules were abundant along the CA3 area in a stratal distribution, with an evident increase compared with NS group (P 〈 0.01 ). In PDTC group, Timm - stained granules were visible, but were markedly reduced in contrast with PTZ seizure group ( P 〈 0.05 ). Meanwhile, in the CA3 area of hippocampus in NS group, a small amount of N -Cadherin positive cells were visible and were slightly stained. Compared with NS group, in PTZ seizure group, N- Cadherin positive cells had a marked increase in area CA3 and dentate hilus ( P 〈 0.01 ), but they were strongly inhibited in PDTC group ( P 〈 0.05 ). There was no correlation between the expressions of N - cadherin, Timm - stained granules and between the age of rats with seizure. Conclusion Single seizure induces mossy fiber sprouting in immature rat hippocampus, and the location and time phase of N - Cadherin are in accord with mossy fiber spouting after single seizure, which suggests N -
