摘 要:目的研究河北省2005-2008年甲3亚型流感病毒HA1基因变异特征。方法选取河北省2005-2008年分离的甲3亚型流感病毒进行核酸提取,采用逆转录-聚合酶链反应(RT-PCR)扩增病毒HA1基因后进行核苷酸序列测定,用生物信息软件对测序结果进行分析处理。结果2005-2006年分离株与当年疫苗株A/California/7/2004比较,在其HA1蛋白分子上有6个氨基酸位点发生替换;2006-2007年分离株与当年疫苗株A/Hiroshima/52/2005比较,有4个氨基酸位点发生替换;2007-2008年分离株与当年疫苗株A/Wisconsin/67/2005比较,有8个氨基酸位点发生替换,且与前两个流行期的分离株比较存在4、6个氨基酸位点差异。基因进化树上,同一流行期分离株基本呈集中分布,每个流行期分离株均远离当年疫苗株。结论河北省2005-2008年分离的甲3亚型流感病毒HA1基因发生了变异,在HA1氨基酸序列的进化亲缘关系结构上具有同流行期相近的特征。Objective To study the genetic variation characteristics of HA1 gene of influenza virus subtype H3N2 isolated in Hebei Province from 2005 to 2008. Methods Viral RNA was extracted and their HA1 genes were amplified by reverse transcription polymerase chain reaction (RT- PCR). The PCR products were sequenced, and the gene sequences were analyzed with the biometric software. Results The HA1 domain of strains isolated in the periods of 2005-2006, 2006-2007, and 2007-2008 took 6, 4 and 8 amino acid substitution sites compared with A/California/7/2004, A/Hiroshima/52/2005 and A/Wisconsin/67/2005 vaccine strains, respectively. There were 4 and 6 amino acid subtitution sites among strains from 2007 to 2008 compared with the strains isolated in the periods of 200.5-2006 and 2006-2007 , respectively. In the phylogenetic tree, the strains of the same year almost clustered in the same group, and the strains isolated from 2005 to 2008 were far away from the current vaccine strain. Conclusions Several from 2005 to 2008, the strains of the same year are very close genetic changes occur in the HA1 domain of the strains isolated in the phylogenetic tree.
