摘  要:毛细管打孔单孢分离是将真菌孢子涂布在2%琼脂板上,显微镜下找到单孢后,用内径1mm毛细管直接打孔,然后挑取带有单孢的琼脂饼培养。毛细管打孔简化了单孢分离的操作过程,提高了单孢分离的速度和成功率,特别适合强寄生真菌的分离培养。用于梨黑星病菌、苹果褐斑病菌和樱桃褐斑病菌的分离培养,成功率达60%以上。Steps to isolate strongly parasitic fungi by single-spore were: smear the fungi spores on to the surface of 2% water agar, find a single-spore under microscope, separate the spore by press a capillary (with the diameter of 1 mm) into the agar, pick the piece of agar wafer with the spore and then transferred to corresponding medium. This method simplified isolation procedures, improved isolation speed and success rate, and was especially suitable for isolating strongly parasitic fungus. The method was used to isolate Venturia nashicola, Diplocarpon mall and Cercospora circumscissa and more than 60% single-spores isolated with this method developed to colonies.
