摘  要:目的了解交感神经递质去甲肾上腺素对肝细胞内核转录因子NFkB p50/p65及其配体IkBα的表达变化的调控。方法应用终浓度为10μmol/L的去甲肾上腺素作用于Wistar大鼠原代培养肝细胞,作用时相点为5、15、30和60min。收集细胞提取细胞核蛋白,采用EMSA法检测NFkB p50/p65的变化。同样以Western blotting法进行Irk3a含量的检测。结果由EMSA电泳图像中可见NE作用5分钟时即可见到有NFkB p50/p65明显活化,并随时间增强而活化逐渐明显。计算灰度面积值在5min后各测得值均较对照组明显增高(P〈0.01)。NE作用肝细胞5min后,IkBa表达量明显减少,刺激30~60min时只能检测到其微量表达(P〈0.01)。结论NE可导致内皮细胞NFkB p50/p65发生核移位,并使IkBα降解,进而活化NFkB,诱导肝细胞分泌细胞因子。Objective To study the effects of norepinephrine (NE) on the activity of nuclear factors in hepatocytes. Methods Primary cultured hepatocytes of Wistar rats were administered with NE( 10 μmol/L) for 5,15, 30 and 60 minutes respectively. The activity of NFkB was measured by EMSA method in each group. Meanwhile, IkB was detected by Western blotting. Results NE increased significantly the levels of NF-kB activity 5 min after administration, and forward gradually at 15, 30 and 60 min(P〈0.01). On the contrary, IkB degraded markedly at 5 min after adminstrtion(P〈0.01), and only minimal activity of IkB could be detected at 30 and 60 min. Conclusion NE might induce the nuclear translocation of hepatocytes NFkB p50/p65 and IkBα degradation and activate NFkB, which ultimately leads to the secretion of cytokines from the hepatocytes.
