摘 要:目的:了解流行性乙型脑炎病毒各蛋白成分在刺激机体免疫应答方面所起的作用,给亚单位疫苗研制提供理论依据,进行该项研究。方法:建立乙脑病毒免疫转印方法,应用该法检查乙脑病人血清IgG抗体对乙脑病毒蛋白的反应情况,分析正常人群中筛选出的乙脑病毒隐性感染者血清IgG对乙脑病毒蛋白的反应性,并比较二者的差异。结果:经检测,绝大部分病人血清IgG可识别E蛋白,部分病人尚可识别NS5,NS3,NS1蛋白,而隐性感染者血清IgG对98kd(NS5)乙脑病毒蛋白的反应性显著低于乙脑患者。结论:研究结果显示,E蛋白是乙脑病人血清IgG识别的主要蛋白,在免疫保护方面起着主要作用。隐性感染者对NS5蛋白的反应机率明显低于显性感染者,提示NS5蛋白在乙脑病毒的致病过程中起重要作用。Objective In order to study the role of Japanese encephalitis virus (JEV) proteins in stimulating immune response of human body,and provide thd theoretical basis for the research of JEV subunit vaccine,we developed a Western blot technique for JEV. METHOD:The specific recognition of antigenic proteins of JEV by sera of 89 patients was investigated by using non-reducing Western immunoblot analysis. The same method was used to detect reaction of 45 sera of inapparent infectioners to JEV proteins. The difference of the two results was compared. RESULTS:We found that the serum IgG antibodies of the most JE patients recognized the E protein,some patients still reacted with NS5,NS3,NS1 protein. The recongization rate of the serum IgG antiboties of JEV inapparent infectioners to JEV NS5 protein was much less than the E protein was Conclusion. CONCLUSION:The results of our study indicated that the E protein was the major protein recognized by JE patients,it played a major role in stimulating immune response. The reconization rate of the serum IgG of inapparent infectioners to NS5 protein was significantly less than that of patients,showing that the NS5 was an important protein in causing disease.
