摘 要:目的本实验系在考察层流剪切力对脐血CD34+细胞体外扩增的影响作用。方法从脐血中分离出CD34+细胞,接种入实验室自制的层流剪切力实验装置中,在不同大小的层流流体剪切力(0.5,1,2,4dyn/cm2)加载下进行体外扩增培养。结果较小的层流剪切力(0.5,1dyn/cm2)可促进造血干/祖细胞扩增,有利于早期造血干/祖细胞的维持,并能促进更多的细胞由G0/G1期进入S期;较大的层流剪切力(2,4dyn/cm2)抑制了造血干/祖细胞的扩增,导致其迅速分化;并使S期细胞的比例减小。结论在生物反应器体外扩增培养脐血CD34+细胞时,其层流流体剪切力应控制在1dyn/cm2范围内。Objective This paper mainly studies the effects of laminar shear stress on the ex vivo expansion of cord blood CD34 + cells. Methods Separate cord blood CD34 + cells, inoculate the cells in the self-made laminar shear stress generation apparatus, and culture the cells under laminar shear stress conditions(0.5,1,2,4 dyn/cm2 ). Results Low laminar shear stress (0.5,1 dyn/cm2 ) had higher expansion folds, better hematopoiesis function, and it can promote more cells from C0/G1 phase to S phase. High laminar shear stress(2,4 dyn/cm2) had negative effects on the ex vivo expansion of hematopoietic stem/progenitor cells, leading to the differentiation, and inhibiting cells from C0/G1 phase to S phase. Conclusion When culturing the CD34 + cells, the fluid laminar shear stress in the bioreactor should he kept below 1 dyn/cm2.
