摘  要:目的:观察更年春方对去卵巢(OVX)大鼠骨髓微环境中骨保护素(OPG)、破骨细胞分化因子(RANKL)和巨噬细胞集落刺激因子(M-CSF)mRNA表达的影响,探讨更年春抗绝经后骨质疏松骨丢失的可能分子机制。方法:取健康10月龄雌性SD大鼠42只,随机平均分为假手术组(S),生理盐水组(N)、尼尔雌醇组(E)、更年春高剂量组(H)、更年春中剂量组(M)和更年春低剂量组(L)。假手术组仅切除卵巢周围少许脂肪,其余5组均切除卵巢。灌药3d后取腰椎冲洗骨髓提取总RNA,实时定量PCR法检测上述因子mRNA的表达情况。结果:更年春高剂量组可上调OVX大鼠骨组织中OPG的mRNA的表达,下调M-CSF的mRNA的表达,使RANKL/OPG值下降。结论:更年春方抗绝经后骨质疏松骨丢失的分子机制可能与其调控OPG和M-CSF的表达,影响RANKL/OPG值有关。Objiective: To observe the effect of Gengnianchun Decoction (GNC) on the mRNA expression of osteoprotegerin(OPG), osteoclast differentiation factor(RANKL) and macrophage colony stimulating factor (M-CSF) in bone marrow microenvironment of ovariectomized rats, and investigate the possible pathway of GNC in preventing and treating postmenopausal osteoporosis. Methods:Forty-two healthy adult SD rats were randomly divided into sham operation group(S group), saline group(N group), Different concentrations of GNC groups: high, moderate and low level groups (H, M, and L group)and nilestriol group (E group). All rats were ovariectomized except those in the sham operation group. After feding with medicines for three days, the total RNA was extracted from the lumbar vertebra to examine mRNA expression of RANKL, OPG and M-CSF by real-time PCR. Results: H group up-regulated of the ovarietomized rat, whereas down-regulated the expression of M-CSF and decreased Conclusion: The effect of GNC in treating postmenopausal osteoporosis is closely correlated with the the expression of OPG RANKL : OPG ratio. regulation of OPG andM-CSF expressions and RANKL/OPG.
