消白合剂对人A375黑素瘤细胞株黑素合成的影响

摘　　要：目的观察消白合剂对人A375黑素瘤细胞生长、酪氨酸酶活性及黑素含量的影响。方法设消白合剂水提液体0.01、0.1、1、10g/L浓度实验组和空白对照组,体外观察对人A375黑素瘤细胞的增殖、酪氨酸酶的活性以及黑素含量的调节作用。四甲基偶氮唑蓝(MTT)比色法测定消白合剂对细胞增殖的影响,酶学方法测定对酪氨酸酶活性的影响,比色法(490 nm)测定对黑素含量的影响。结果消白合剂在0.01～0.1 g/L浓度范围内对细胞增殖、酪氨酸酶活性及黑素的合成有促进作用,而随着浓度的继续增高反而起抑制作用。结论消白合剂对人A375黑素瘤细胞株黑素的合成具有双向调节作用,临床运用时应该考虑到药物的量的优化。Objective To observe the effects of ＂Xiaobai Mixture＂ on the growth of human melanoma cell line A375, the activity of tyrosinase and the amount of Melanin. Methods The human melanoma cell A375 was divided into experimental groups in terms of various concentrations（0.01,0.1, 1, 10g/L） of ＂Xiaobai Mixture＂ and control group. Then the proliferation of melanocytes, tyrosinase activity and melanic concentration were observed in vitro. MTT method was used to detect the proliferation of melanocytes; the tyrosinase activity of melanocytes was observed by zymologic method; the amount of melanin was observed by colorimetry （490nm）. Results The proliferation of melanocytes, tyrosinase activity and melanic concentration were significantly increased by the treatment with ＂Xiaobai Mixture＂ at the concentrations of 0.01 - 10g/L. But it has inhibitory action when the concentration was continuously increased. Conclusion ＂Xiaobai Mixture＂ has bidirectional adjusting action in the growth of human melanoma cell line A375, suggesting that the dosage of ＂Xiaobai Mixture＂ should be optimized in clinical application.
