摘 要:目的探讨三丁基锡的肥胖激素效应。方法将幼龄非洲爪蟾(Xenopus laevis)置于浓度分别为50和100 ng/L的氯化三丁基锡溶液中暴露3个月,检测非洲爪蟾的体重、体长、躯干面积和脂肪重量等生长指标和肝脏组织的脂质变化。结果暴露1个月后,50 ng/L组体重比对照组增加了0.7倍,100 ng/L组体重、体长、躯干面积和脂肪重量比对照组分别增加了1.6、0.5、1.5和1.8倍;暴露2个月后,50 ng/L组躯干面积比对照组增加了0.5倍,100 ng/L组体重、体长、躯干面积和脂肪重量比对照组分别增加了0.6、0.2、0.7和2.8倍;暴露3个月后,100 ng/L组体长、躯干面积和脂肪重量比对照组分别增加了0.2、0.5和0.6倍。冰冻切片的脂质染色发现,暴露3个月,100 ng/L组中肝脏脂肪积累面积比对照组增加了1.7倍。结论TBT能够引起非洲爪蟾体重、体长、躯干面积和脂肪重量的增加,造成肝脏脂肪的积累,具有环境肥胖激素效应。鉴于非洲爪蟾在甲状腺激素和性激素等环境内分泌干扰物筛选中的广泛应用,可以考虑将其作为筛选环境肥胖激素的实验动物模型。Objective To explore the obesogen effects of tributyltin on vertebrates. Methods Africa clawed frogs (Xenopus laevis) were exposed to the water containing 50 and 100 ng/L tributyhin chloride for 3 consecutive months. The changes of growth index and accumulation of lipoid in the liver were observed. Results After one month of exposure, the body weight (BW) in 50 ng/L tributyhin exposure group increased by 0.7 times compared with the control, while BW, body length (BL), trunk area (TA) and fat weight (FW) in 100 ng/L tfibutyltin exposure group increased by 1.6, 0.5, 1.5 and 1.8 times respectively compared with the control. After two months of exposure, TA in 50 ng/L tributyltin exposure group elevated to 1.5 fold of that in the control, while BW, BL, TA and FW in 100 ng/L group was 1.6, 1.2, 1.7 and 3.8 fold of those in the control, respectively. After three months of exposure, BL, TA and FW in 100 ng/L treatment group was 1.2, 1.5 and 1.6 fold of those in the control. In addition, TBT exposure induced the accumulation of fat droplets in the liver and the fatty degree in 100 ng/L group reached 2.7 fold of that in the control after 3 months of exposure. Conclusion The results of the present paper suggest that TBT can induce the obesogen effect on Xenopus laevis, which can serve as the animal model to study the obesogen effects since it has been widely used in screening thyroid and sex hormones disrupting chemicals.
