摘 要:目的研究哺乳动物雷帕霉素靶蛋白mTOR特异性抑制剂雷帕霉素(Rapamycin)对体外培养的人结肠癌Lovo细胞生长的抑制作用,并初步探讨其机制。方法分别用0.5、1.0、2.5和5.0μg/mL雷帕霉素对Lovo细胞进行干预,采用MTT检测细胞增殖,流式细胞术检测细胞凋亡,Westernblot检测凋亡抑制蛋白Bcl-2的表达。结果雷帕霉素对Lovo细胞表现出明显的增殖抑制作用,细胞增殖活性随浓度增加、时间延长逐渐降低,呈现量-效、时-效关系。雷帕霉素呈浓度依赖性诱导细胞凋亡,并明显降低Lovo细胞中凋亡抑制蛋白Bcl-2的表达水平。结论雷帕霉素通过下调凋亡抑制蛋白Bcl-2的表达,诱导结肠癌Lovo细胞凋亡,从而抑制其增殖及生长。[Objective] To investigate the effect of Rapamycin, a inhibitor of the mTOR, on the viability of the colon carcinoma cell line Lovo. [Methods] The cultured Lovo cells were treated with different concentrations of Rapamycin (0.5, 1.0, 2.5 and 5.0 μg/ml) for different time. The proliferation of Lovo cells was detected by MTT method. Cell apoptosis was detected by flow cytometry. The levels of Bcl-2 protein were determined by Western blot. [Resuits] Rapamycin inhibited the proliferation of Lovo cells in a concentration- and timedependent manner. Rapamycin induced the apoptosis of Lovo cells in a concentration-dependent manner. The levels of Bcl-2 protein were reduced gradually with the increase of concentration or action time. [ Conclusion] Rapamycin can induce the apoptosis of the colon carcinoma cell line Lovo, reduce the expression of Bcl-2, and inhibite the proliferation of colon carcinoma.
