摘  要:目的观察两种丝裂原蛋白激酶(mitogen activated protein kinases,MAPK)抑制剂PD98059及U0126对刚地弓形虫侵入宿主细胞的影响,探讨其对弓形虫速殖子侵入宿主细胞信号转导途径的不同阻断效应。方法丝裂原蛋白激酶抑制剂PD98059或U0126分别在不同时间及不同剂量作用于速殖子-宿主细胞培养系统,用流式细胞仪(FCM)检测宿主细胞感染速殖子的差异。结果流式细胞仪检测加入U01261μmol/L、10μmol/L和100μmol/L的细胞培养孔的细胞感染弓形虫速殖子的量分别比对照组平均降低了26.10%(P<0.01),66.42%(P<0.01)和70.39%(P<0.01)。而加入PD980591μmol/L、10μmol/L和100μmol/L的分别比对照组平均降低了25.45%(P<0.01),53.01%(P<0.01)和64.70%(P<0.01)。实验中发现100μmol/L U0126作用培养细胞9h的时候,可导致HL-60细胞出现部分聚集成团、漂浮的中毒现象。结论U0126和PD98059均可明显抑制弓形虫速殖子侵入宿主细胞,但其差异无显著性,其机制有待进一步探索。To observe the effect of the different mitogen activated protein kinases(MAPK) inhibitor on Toxoplasma gondii taehyzoites invading to host cells,the MAPK inhibitor was added to the tachyzoite-HL-60 cell culture in different dosage, then the infected cells were counted in different time(3-9h) by flow cytometry(FCM). The result showed that the rate of HL-60 cells infected by T. gondii tachyzoites was lower remarkably than that of the control group, with 1μmol/L, 10μmol/L and 100μmol/L of U0126, the average infecting rates was decreased to 26.10%, 66. 420/60 and 70. 390% respectively(P〈0.01). With 1μmol/L,10μmol/L and 100μmol/L of PD98059, the average infective rates was decreased to 25.45%, 53. 01% and 64. 70% respectively(P〈0.01). But 100μmol/L U0126 can cause the HL-60 cell to poison at 9 hours. U0126 or PD98059 can remarkably inhibit T. gondii tachyzoites invading to ceils. The inhibiting effect of U0126 was higher than PD98059 but the difference showed no statistic significance.
