摘 要:目的:研究多发性肌炎(PM)和皮肌炎(DM)外周血单一核细胞(PBMCs)中CD45RA^+、CD45RO^+细胞亚群的变化。方法:应用双色荧光标记抗体、流式细胞仪对PM和DM外周血CD45RA^+、CD45RO^+细胞亚群进行了检测。结果:(1)PM和DM的PBMCs中CD45RA^+细胞显著低于对照组,CD45RO^+细胞和CD45RO^+/RA^+比值均显著高于对照组,PM的CD45RA^+细胞显著低于DM(均P〈0.05)。(2)在PM的CD4^+亚群中,CD45RA^+细胞显著低于对照组和DM,CD45RO^+细胞和CD45RO^+/RA^+比值显著高于对照组(均P〈0.05)。(3)在DM的CD4^+亚群中,CD45RA^+细胞与对照组比较无显著性差异(P〉0.05),CD45RO^+细胞和CD45RO^+/RA^+比值显著高于对照组(均P〈0.05)。(4)在CD8^+亚群中,PM和DM的CD45RA^+、CD45RO^+细胞与对照组比较,均无显著性差异(均P〉0.05),但CIM5RO^+/RA^+比值均显著高于对照组(均P〈0.05)。结论:在PM和DM自身免疫发病过程中PBMCs和CD4^+、CD8^+亚群中的CD45RO^+、CD45RA^+细胞可能起重要作用,PM和DM在免疫学方面存在一定的异质性。Objective: To investigate the changes of CIMSRA^+/CIMSRO^+ cell subsets in peripheral blood mono- nuclear cells (PBMCs) in polymyositis ( PM )/dermatomyositis ( DM ). Methods: The changes of CD45RA^+ and CD45RO^+ cell subsets in peripheral blood of PM/DM were detected by two color flourescence labeled antibody and flow cytometry. Results: (1) The CD45RA^+ cells in PBMCs in PM and DM were decreased significantly than that in control group and the CD45RO^+ cells and the ratio of CD45RO^+/CD45RA^+ were both significantly higher as compared to control group. The number of CD45RA^+ cells were significantly lower in PM than in DM ( P 〈 0.05). (2) In PM, the number of CD45RO^+ cells was significantly lower than in control group and in DM. The number of CD45RO^+ cells and the ratio of CD45RA^+/CD45RAO^+ were both significantly higher than that in control group ( P 〈 0.05). (3) In DM, the CD45RA^+ cells was not significantly defferent from that in control group ( P 〉 0.05), and the number of CD45RO^+ cells and the ratio of CD45RO^+/CD45RA^+ were significantly higher than that in con- trol group ( P 〈 0.05). (4) In CD8^+ subset, the CD45RA^+ cells and CD45RO^+ cells in PM and DM were not significantly defferent from that in control group ( P 〉 0.05), but the ratio of CD45RO^+/CD45RA^+ in PM and DM was significantly higher than that in control group (P〈0.05). Conclusion: The rising of CD45RO^+ cells in PBMCs, and the CD45RO^+/CD45RA^+ cells in CD4^+ and CD8^+ subsets may play an important role in the pathogenesis in PM and DM, which may present heterogeneity in immunology.
