摘 要:采用不同剂量(0~10 Gy)60Coγ射线照射人外周血淋巴细胞,通过抽提mRNA,采用real-timePCR检测不同剂量照射后淋巴细胞中GADD45和CDKN1A基因mRNA表达水平的变化。筛选出最佳照射剂量(5 Gy)后,用该最佳照射剂量照射淋巴细胞,检测不同时间点(0~72 h)淋巴细胞中GADD45和CDKN1A基因mRNA表达水平的变化。结果表明,淋巴细胞中GADD45和CDKN1A基因mRNA的表达水平随照射剂量的增加而增加,并呈现出剂量效应关系。在5 Gy最佳剂量照射后,淋巴细胞GADD45基因mRNA表达水平随时间的推移不断增高,在8 h时达到最高,随后开始逐渐降低;淋巴细胞CDKN1A基因mRNA表达水平随时间的推移未见明显变化趋势。淋巴细胞系中GADD45和CDKN1A基因mRNA的表达水平与照射剂量均有较好的剂量效应关系,两者均有可能作为电离辐射的生物剂量计来估算受照剂量水平。This study is to investigate the changes of GADIM5 and CDKN1A gene mRNA expression induced by ^60Coγ-ray in lymphoblastoid cell line in vitro, and its probability of gene expression as a biodosimeter. Lymphoblastoid cell line was irradiated by ^60Coγ-ray with the dose of 0 - 10 Gy and cultivated for 8 hours, in order to screen the optimal dose. mRNA was extracted and real-time PCR method was used to detect the changes of PIG3 and GADIM5 gene mRNA expression in lymphoblastoid cells. Lymphoblastoid cells was cultivated for 0 -72 h after optimal dose irradiation. The changes of GADD45 and CDKNIA gene mRNA expression in lymphoblastoid cells in different time-point were detected. The resuits showed that GADD45 and CDKN1A gene expression in lymphoblastoid cells increased in a dose-dependent form. The peak expression of GADD45 was at 8 h after 5 Gy irradiation and began to decrease. The trend in change of CDKN1A gene expression was not notable.
